      摘 要:[目的]探讨RNA干扰NF-κBp65基因表达对胃癌细胞株SGC-790增殖、凋亡的影响。[方法] 培养SGC-790胃癌细胞株,分为阴性对照组、siRNA-NC组、NF-κBp65-siRNA转染组。检测不同组细胞增殖抑制率及凋亡率,采用Western blot检测STAT3信号通路相关蛋白表达水平。[结果] (1)MTT检测发现,在48h及72h时,NF-κBp65-siRNA组的细胞抑制率显著高于空白组与siRNA-NC组(P均<0.05)。(2)流式细胞术检测发现,48h时,空白组、siRNA-NC组、NF-κBp65-siRNA组的细胞凋亡率分别为5.85%±0.25%、6.13%±0.39%、16.05%±0.14%,NF-κBp65-siRNA组的细胞凋亡率显著增加(P<0.001)。(3)Transwell检测结果显示,空白组、siRNA-NC组、NF-κBp65-siRNA组穿过Transwell小室的细胞数量分别为379.62±10.51、386.86±11.42、89.62±5.73,NF-κBp65-siRNA组的穿膜细胞数量显著少于空白组与siRNA-NC组(P均<0.001)。(4)Western-blot检测显示,与空白组相比,NF-κBp65-siRNA组中p-STAT3、PCNA、MMP-2蛋白的表达量均显著下降(P均<0.05)。[结论]在SGC-790胃癌细胞株中,抑制NF-κBp65表达水平可能通过降低STAT3信号通路活性,抑制细胞增殖并促进凋亡。

      Abstract:[Objective] To investigate the effect of RNA interference of NF-κBp65 on proliferation and apoptosis of gastric cancer cell line SGC-790. [Methods] SGC-790 gastric cancer cells were divided into control group,siRNA-NC group and NF-κBp65-siRNA group. Cell proliferation inhibition rate and apoptosis rate were determined with MTT assay and cytometry,respectively;cell migration was examined with Transwell test;and the expression of STAT3 signaling pathway related proteins were detected by Western blot. [Results] MTT assay showed that the inhibition rate of SGC-790 cells in NF-κBp65-siRNA group was significantly higher than that in control group and siRNA NC group at 48 h and 72 h(all P<0.05). Flow cytometry showed that the apoptosis rate in NF-κBp65-siRNA group was significantly higher than those in control group and siRNA-NC group(16.05%±0.14% vs 5.85%±0.25% and 6.13%±0.39%,P<0.001). The Transwell test results showed that the number of passing cells in NF-κBp65-siRNA group was significantly less than that in control group and siRNA-NC group(89.62±5.73 vs 379.62±10.51 and 386.86±11.42,P<0.001). Compared with the control group,the expression of p-STAT3,PCNA and MMP-2 protein in NF-κBp65-siRNA group decreased significantly(all P<0.05). [Conclusion] The knockdown of NF-κBp65 can inhibit cell proliferation and promote apoptosis in SGC-790 gastric cancer cells,which may be associated with down-regulation of STAT3 signaling pathway activity.
