      摘 要:[目的] 研究食管鳞状细胞癌组织中miR-34a表达水平及其临床意义。[方法] 采用实时反转录聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)技术检测75例食管鳞状细胞癌组织及配对癌旁组织中miR-34a的表达水平,2-ΔΔCt法计算食管鳞状细胞癌组织中miR-34a相对表达量,进一步结合临床、病理及随访资料进行统计分析。[结果] miR-34a在75例食管鳞癌组织中相对表达量高于癌旁组织;多因素分析显示,吸烟史和临床分期是食管鳞癌组织中miR-34a表达的独立影响因素。miR-34a低表达组的无进展生存期和总生存期均明显劣于高表达组;多因素分析显示miR-34a低表达是无进展生存期和总生存期缩短的独立影响因素。[结论] 食管鳞癌组织中miR-34a表达水平可作为预测预后的一项重要指标。

      Abstract:[Purpose] To investigate the expression of miR-34a in esophageal squamous cell carcinoma(ESCC) and its clinical significance. [Methods] The expression of miR-34a in 75 cases with ESCC and adjacent cancer tissues was tested by reverse transcription-polymerase chain reaction(RT-PCR). 2-ΔΔCt method was used to calculate the relative quantitative expression of miR-34a,which was analyzed together with clinicopathological and follow-up data. [Results] The expression of miR-34a was higher in the tumor tissues than that in cancer adjacent tissues. Multivariate analysis showed that smoking and clinical stage were independent factors influencing the expression of miR-34a in ESCC.Progression-free survival and overall survival in lower miR-34a expression group were worse than those in higher miR-34a expression group. Multivariate analysis indicated that lower miR-34a expression was independently associated with poorer progression-free survival and overall survival. [Conclusions] MiR-34a might be an important indicator for prediction of prognosis of ESCC.
