      摘 要:[目的] 通过体外培养和流式细胞术,检测贝母甲素对人急性髓系白血病细胞KG-1a增殖抑制、诱导凋亡和细胞周期分布的影响。[方法] 利用MTT法检测贝母甲素对KG-1a的增殖抑制作用,计算抑制率;以流式细胞术检测贝母甲素对细胞凋亡及周期分布的影响。[结果] ①与对照组比较,贝母甲素作用于KG-1a细胞24h、48h、72h的吸光度(OD)值均明显降低(P<0.05),且细胞生长抑制率与药物浓度呈正相关。②与对照组比较,贝母甲素能够提高KG-1a细胞晚期凋亡及总凋亡率,差异均有统计学意义(P=0.003,P=0.001)。③贝母甲素干预KG-1a后,细胞周期发生明显变化,G0/G1期细胞比例降低、G2/M期细胞比例升高、凋亡率升高(P=0.001,P=0.002,P=0.046)。[结论] 贝母甲素具有抑制具有LSC特性的KG-1a细胞增殖、降低G0/G1期细胞比例、升高G2/M期细胞比例、诱导细胞凋亡效应。

      Abstract:[Purpose] To detect the influence of Verticine on human acute myeloid leukemia cell KG-1a in inhibition proliferation,induction apoptosis and cell cycles by flow cytometry in vitro. [Methods] The effect and ratio of inhibition proliferation were conducted by MTT method. Cell apoptosis and the cell cycle distribution were detected by flow cytometry. [Results] (1) Compared with the control group,the absorbance(OD) values of KG-1a cells at 24 h,48 h and 72 h in Verticine group decreased(P<0.05). The inhibition rate of cell growth was positively correlated with increased drug concentration.(2) Verticine improved the late apoptosis and apoptotic rate of KG-1a cells,which were significantly different from control group(P=0.003,0.007). (3)After Verticine intervention,the cell cycle of KG-1a cells changed significantly:the value of G0/G1 phase declining,along with G2/M phase and cell apoptosis rate increasing(P=0.001,0.002,and 0.046). [Conclusion] Verticine can inhibit LSC KG-1a cell proliferation,enhance the G2/M phase ratio,decrease G0/G1 phase ratio and induce cell apoptosis.
