| 摘 要：叉头框蛋白O3a （forkhead box class O3a，FOXO3a）是叉头框蛋白O家族的重要转录因子，作为磷脂酰肌醇3-激酶（phosphoinositide 3-kinase，PI3K）/蛋白激酶B（protein kinase B，AKT）信号通路下游关键的转录因子，通过调节细胞周期抑制因子、促凋亡基因及抗氧化应激相关基因的转录，在维持细胞稳态、抑制肿瘤细胞异常增殖和调控细胞炎症反应中发挥关键作用。FOXO3a在不同肺癌组织学类型的发生及进展过程中具有重要调控意义，FOXO3a普遍表现为抑制肿瘤生长、侵袭及转移的抑癌因子，其表达下调或功能失活与肿瘤进展、侵袭转移及治疗耐药性相关。全文总结分析FOXO3a参与调控非小细胞肺癌和小细胞肺癌发生及进展的分子机制，为肺癌的诊断、预后评估及免疫靶向治疗提供新的理论依据和潜在分子靶点。 |

| Abstract： Forkhead box class O3a(FOXO3a)， a key transcription factor of the forkhead box O family， acts downstream of the phosphoinositde 3-kinase (PI3K)/protein kinase B (AKT) signaling pathway. It regulates the transcription of cell cycle inhibitors， pro-apoptotic genes， and antioxidant stress related genes， thereby playing a critical role in maintaining cellular homeostasis， inhibiting abnormal tumor cell proliferation， and modulating inflammatory responses. FOXO3a exhibits significant regulatory functions in the carcinogenesis and progression of various pathological types of lung cancer. It generally functions as a tumor suppressor， inhibiting tumor growth， invasion， and metastasis. Downregulation or functional inactivation of FOXO3a is associated with tumor progression， metastasis， and therapeutic resistance. This paper reviews and analyzes the molecular mechanisms by which FOXO3a regulates the carcinogenesis and progression of non-small cell lung cancer and small cell lung cancer， providing a new theoretical basis and potential molecular targets for the diagnosis， prognosis assessment， and immunotargeted therapy of lung cancer. |
