| 摘 要:[目的] 探讨PPT2在消化道肿瘤中的差异表达及作用。[方法] 对癌症基因组图谱(The Cancer Genome Atlas,TCGA)和GTEx数据库中的胃癌、食管癌、结肠癌、直肠癌以及癌旁组织数据集进行分析。采用R包 limma进行差异分析。基于Metascape对差异表达基因进行富集分析。使用R包survival进行生存分析。使用STRING数据库构建蛋白质互作(protein-protein interaction,PPI)网络。最后,通过蛋白质印迹分析(Western blot analysis)验证蛋白表达。[结果] 在4种消化道肿瘤中具有大量共同差异表达基因,这些基因与肿瘤的各个生物功能及预后密切相关,最终筛选出在4种消化道肿瘤组织中均低表达的棕榈酰蛋白硫酯酶2(palmitoyl protein thioesterase,PPT2)基因。生存分析结果显示,在食管癌和直肠癌患者中PPT2低表达者预后较差,而在结肠癌和胃癌中PPT2高表达患者预后较差(P均<0.05)。对PPT2进行PPI分析,并对PPI网络的基因进行富集分析;基因本体论(Gene Ontology,GO)分析显示PPT2与脂肪酸-β氧化、溶酶体组织、脂肪酸生物合成过程等有关;京都基因和基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)通路分析显示PPT2与脂肪酸生物合成、酒精性肝病有关。Western blot验证结果显示,PPT2在结肠癌、食管癌、胃癌及直肠癌组织中的表达均低于癌旁组织(P均<0.05)。[结论] 胃癌、食管癌、结肠癌及直肠癌具有大量共同的差异表达基因,其中PPT2在4种肿瘤中显著差异表达且与预后相关,是潜在的重要生物标志物及治疗靶点。 |

| Abstract: [Objective] To investigate the differential expression of PPT2 in gastrointestinal cancers and its prognostic significance. [Methods] Datasets of gastric cancer, esophageal cancer , colon cancer, rectal cancer, and paracancerous tissues were obtained from The Cancer Genome Atlas (TCGA) and GTEx databases. Differential expression analysis was performed by the R package limma. Enrichment analysis of differentially expressed genes was conducted by Metascape. Survival analysis was performed by the R package “survival”. The protein-protein interaction (PPI) network was constructed by the STRING database. Finally, PPT2 expression was validated by Western blot analysis. [Results] A large number of common differentially expressed genes were identified across the four gastrointestinal cancer types, closely associated with tumor biological functions and prognosis. Among these, palmitoyl protein thioesterase 2 (PPT2) was consistently down-regulated in all four tumor tissues compared to paracancerous tissues. Survival analysis revealed that patients with low PPT2 expression had a poorer prognosis in esophageal cancer and rectal cancer, whereas high PPT2 expression was associated with worse prognosis in colon cancer and gastric cancer (all P<0.05). PPI analysis of PPT2 and enrichment analysis of its interacting genes showed that PPT2 was significantly enriched in biological processes such as fatty acid beta-oxidation, lysosome organization, and fatty acid biosynthetic process (GO analysis). Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis indicated associations with fatty acid biosynthesis and alcoholic liver disease. Western blot results confirmed that PPT2 protein expression was significantly lower in colon cancer,esophageal cancer, gastric cancer, and rectal cancer tissues than in paracancerous tissues(P<0.05). [Conclusion] Gastric cancer, esophageal cancer, colon cancer and rectal cancer share a substantial number of common differentially expressed genes. PPT2 is significantly differentially expressed across these four gastrointestinal cancers and is associated with prognosis, suggesting its potential as a biomarker and a therapeutic target. |
