      摘 要:[目的] 探讨免疫球蛋白超家族成员9(immunoglobulin superfamily member 9,IGSF9)及缺氧诱导因子-1α(hypoxia-inducible factor-1α,HIF-1α)在子宫内膜癌(endometrial cancer,EC)中的表达及其与预后的相关性。[方法] 选取石家庄市第四医院EC患者82例,同期子宫内膜不典型增生(endometrial intraepithelial neoplasia,EIN)患者54例,癌旁正常子宫内膜组织52例。采用免疫组织化学染色及实时荧光定量聚合酶链式反应检测IGSF9、HIF-1α蛋白及IGSF9、HIF-1α mRNA表达,Kaplan-Meier法绘制生存曲线, Cox回归分析IGSF9、HIF-1α蛋白与EC患者临床病理特征及预后关系。[结果] 在EC、EIN及正常子宫内膜中,IGSF9蛋白阳性表达率分别为56.10%、35.18%、23.08%,HIF-1α蛋白阳性表达率分别为68.29%、44.44%、34.61%,差异均具有统计学意义(IGSF9:χ2=15.392,P<0.001;HIF-1α:χ2=16.254,P<0.001)。EC组织中IGSF9、HIF-1α蛋白表达与淋巴结转移、FIGO分期、肌层浸润深度相关(P均<0.05)。IGSF9及HIF-1α mRNA在EC中表达水平均高于EIN及正常子宫内膜(P均<0.01)。多因素Cox回归分析表明IGSF9表达是EC患者预后差的独立影响因素(HR=3.279,95%CI:1.096~9.817)。IGSF9阳性EC患者5年总生存率低于阴性者(50.0% vs 88.9%,χ2=11.438,P=0.001)。HIF-1α阳性EC患者5年总生存率也低于阴性者,但差异无统计学意义(62.5% vs 76.9%,χ2=3.424,P=0.064)。[结论] IGSF9及HIF-1α在EC中高表达,可能共同参与EC进展及转移。

      Abstract: [Objective] To investigate the expression of hypoxia-inducible factor-1α (HIF-1α) and immunoglobulin superfamily member 9 (IGSF9), and their association with prognosis in endometrial carcinoma (EC). [Methods] A total of 82 EC, 54 endometrial intraepithelial neoplasia (EIN), and 52 normal endometrial tissue were collected from The Fourth Hospital of Shijiazhuang. Immunohistochemistry and quantitative real-time polymerase chain reaction were used to detect the expression of HIF-1α and IGSF9 at the protein and mRNA levels, respectively. Survival curves were plotted using the Kaplan-Meier method, and Cox regression analysis was conducted to explore the relationship between IGSF9 and HIF-1α expression and the clinicopathological characteristics and prognosis of EC patients. [Results] The positive expression rates of IGSF9 protein in EC, EIN and normal endometrium were 56.10%, 35.18% and 23.08%, respectively, and the positive expression rates of HIF-1α protein were 68.29%, 44.44% and 34.61%, respectively. The differences were statistically significant (IGSF9: χ2=15.392, P<0.001; HIF-1α: χ2=16.254, P<0.001). The expression of IGSF9 and HIF-1α expression in EC tissues were related to lymph node metastasis, FIGO stage and infiltrative depth (all P<0.05). The expression levels of IGSF9 and HIF-1α mRNA in EC were both higher than those in EIN and normal endometrium (all P<0.01). Multivariate Cox regression analysis showed that the IGSF9 expression was an independent influencing factor for poorly prognosis in EC patients(HR=3.279, 95%CI: 1.096~9.817). The 5-year overall survival of IGSF9-positive EC patients was lower than that of the negative group (50.0% vs 88.9%, χ2=11.438, P=0.001). The 5-year overall survival of HIF-1α-positive EC patients was also lower than that of the negative group, but the difference was not statistically significant (62.5% vs 76.9%, χ2=3.424, P=0.064). [Conclusion] HIF-1α and IGSF9 are upregulated in EC, and may jointly contribute to disease progression and metastasis.
