摘 要:[目的] 分析泛癌中的COL5A1基因及蛋白表达水平,并探讨其与临床预后和肿瘤免疫浸润的关系。[方法] 分析癌症基因组图谱(The Cancer Genome Atlas,TCGA)数据库中肿瘤患者的数据,采用TIMER2.0、GEPIA2、SangerBox、人类蛋白图谱(human protein atlas,HPA)和STRING分析等生物信息学工具分析COL5A1基因与泛癌的关系。[结果] COL5A1在17种不同肿瘤和癌旁组织中的表达水平有差异。COL5A1高表达与膀胱尿路上皮癌、肺鳞状细胞癌、肺腺癌、宫颈鳞状细胞癌和宫颈腺癌、甲状腺癌、卵巢癌、肾透明细胞癌、肾乳头状细胞癌和胃腺癌总生存期相关(P均<0.05)。COL5A1表达与肿瘤微环境免疫检查点、肿瘤突变负荷和微卫星不稳定性显著性相关。COL5A1表达与B细胞、T细胞、中性粒细胞、巨噬细胞和抗原递呈细胞相关。[结论] COL5A1是肿瘤微环境的重要组成部分,影响免疫细胞浸润,可作为评估多种恶性肿瘤的免疫浸润和预后分析的肿瘤标志物。 |

Abstract:[Objective] To investigate the gene and protein expression levels of the COL5A1 in pan-cancer and its biological and clinical implication. [Methods] Data from cancer patients were collected from The Cancer Genome Atlas (TCGA) database, and its biological and clinical implication in pan-cancer were analyzed with TIMER2.0, GEPIA2, SangerBox, human protein atlas (HPA) and STRING. [Results] The gene and protein expression levels of COL5A1 varied in 17 different tumors and adjacent tissues. High expression of COL5A1 was significantly associated with shorter overall survival in bladder urothelial carcinoma, lung squamous cell carcinoma, lung adenocarcinoma, cervical squamous cell carcinoma and cervical adenocarcinoma, thyroid cancer, ovarian cancer, clear cell renal carcinoma, papillary renal carcinoma, and gastric adenocarcinoma (all P<0.05). COL5A1 expression was significantly correlated with immune checkpoints in the tumor microenvironment, tumor mutation burden, and microsatellite instability in human cancers. Positive expression of COL5A1 was associated with immune infiltration of B cells, T cells, neutrophils, macrophages, and antigen-presenting cells in tumors. [Conclusion] COL5A1 is an important component of the tumor microenvironment, influencing immune cell infiltration, and it can serve as a tumor marker for assessing immune infiltration and prognosis in various malignant tumors. |
