摘 要: 肿瘤微环境在人类实体肿瘤的恶性侵袭行为中发挥着关键作用。在与肿瘤微环境相关的细胞类型中,大多数实体瘤的骨髓浸润中肿瘤相关巨噬细胞(tumor-associated macrophages,TAMs)及其前体细胞占最大比例,甚至可以占肿瘤质量的50%。由于TAMs具有M2型巨噬细胞的大多数特性,TAMs通常被认为是M2型的巨噬细胞。TAMs作为免疫抑制细胞中重要的一员,能够通过分泌多种因子对肿瘤微环境产生广泛的影响,进而在肿瘤发生及进展中发挥关键作用。全文主要从TAMs的起源、巨噬细胞的极化和表型转换出发,讨论TAMs各种促肿瘤功能,包括肿瘤的增殖、侵袭和迁移、促进血管和淋巴管生成以及介导免疫抑制等,以及与巨噬细胞相关的治疗策略,并总结了TAMs在肾透明细胞癌、非小细胞肺癌、肝癌、甲状腺癌、乳腺癌中的国内外研究动态。 |

Abstract: Tumor microenvironment (TME) plays a crucial role in the malignant invasion behavior of human solid tumors. Among all cell types in TME, tumor-associated macrophages(TAMs) and their precursors account for the largest proportion, even for 50% of the tumor mass of most solid tumors. TAMs possesses most features of M2 macrophages, so it is generally considered to be M2 macrophages. Meanwhile, as an important member of immunosuppressive cells, TAMs exert a wide range of effects on TME by secreting various factors, thereby playing a crucial role in tumor occurrence and development. The paper reviews the origin of TAMs, polarization of macrophages and phenotype transformation, elaborates its pro-tumor functions, including promoting tumor proliferation, invasion and migration, promoting angiogenesis and lymphangiogenesis, and mediating immune suppression. The paper also discusses TAMs-related treatment strategies for clear cell renal cell carcinoma, non-small cell lung cancer, liver cancer, thyroid cancer and breast cancer. |
