摘 要:[目的] 探讨蒲公英提取物(dandelion extract,DE)对胃癌细胞增殖、凋亡、周期、迁移及侵袭的影响,并探讨其分子机制。[方法] 通过蒸馏和冷冻干燥技术制备DE,并将不同浓度(0.280、0.560、1.125 μmol/L)的DE分别作用于BGC-823和AGS胃癌细胞株。采用MTS法、Transwell实验、流式细胞术检测各组细胞的增殖、凋亡、周期、迁移及侵袭,采用逆转录定量聚合酶链反应(RT-qPCR)和蛋白质印迹实验(Western blotting)分别检测增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)、B细胞淋巴瘤-xl(B-cell lymphoma extra-large,Bcl-xl)、B细胞淋巴瘤放大因子2(B-cell lymphoma-2,Bcl-2)、基质金属蛋白酶9 (matrix metalloprotein 9,MMP9)、E-钙黏附素(E-cadherin)、白细胞介素(Interleukin,IL)-6、IL-8和IL-1β的表达。[结果] 与对照组相比,DE处理组的细胞活力呈时间和浓度依赖性下降(P均<0.05),细胞凋亡率随着DE浓度的增加而显著增加(P<0.05)。DE处理组的细胞在G0/G1期所占比例最高,而在G2/M期所占比例最低(P<0.05)。与对照组相比,DE处理组的细胞迁移能力和侵袭能力随着DE浓度的增加而逐渐降低(P均<0.05),PCNA、Bcl-xl、Bcl-2、MMP9、E-cadherin、IL-6、IL-8和IL-1β的蛋白和mRNA表达水平均显著降低(P<0.05)。[结论] DE可抑制胃癌细胞增殖、迁移和侵袭,促进细胞凋亡,诱导细胞周期阻滞,可能与其调控相关基因和蛋白的表达有关。 |

Abstract:[Objective] To investigate the effects of dandelion extract (DE) on proliferation and biological activities of gastric cancer (GC) cells and its molecular mechanism. [Methods] DE was prepared by distillation and freeze-drying techniques, and gastric cancer BGC-823 and AGS cells were treated with different concentrations (0.280, 0.560, 1.125 μmol/L) of DE. The proliferation, apoptosis, cell cycle, migration, and invasion of gastric cancer cells by MTS method, Transwell assay, and flow cytometry. The mRNA and protein expression of proliferating cell nuclear antigen (PCNA), B-cell lymphoma extra-large (Bcl-xl), B-cell lymphoma-2 (Bcl-2), matrix metalloproteinase 9 (MMP-9), E-cadherin, IL-6, IL-8 and IL-1 β were detected by RT-qPCR and Western blotting, respectively. [Results] Compared with the control group, the proliferation of the DE treated groups decreased in a time and concentration-dependent manner (all P<0.05), and the apoptosis rate increased significantly with the increase of DE concentration (P<0.05). The proportion of G0/G1 phase cells was higher and G2/M phase cells was lower in the DE treated groups (all P<0.05). Compared with the control group, the migration and invasion abilities of cells in the DE treated groups decreased with increasing DE concentration (P<0.05), and the protein and mRNA expression of PCNA, Bcl-xl, Bcl-2, MMP9, E-cadherin, IL-6, IL-8 and IL-1 β were significantly reduced (all P<0.05). [Conclusion] DE can inhibit the proliferation, migration and invasion of gastric cancer cells, promote cell apoptosis and induce cell cycle arrest, which may be related to its regulation of the expression of related genes and proteins. |
