      摘 要:[目的] 探讨结肠癌患者癌组织中赖氨酸羟化酶2(PLOD2)、巨噬细胞表达共刺激分子VSIG4表达及意义。[方法] 收集经病理确认的Ⅰ~Ⅳ期结肠癌患者,应用免疫组化方法检测癌组织及其癌旁组织中的PLOD2、VSIG4表达,分析两者表达的相关性及与患者临床病理特征的关系。随访并探讨PLOD2、VSIG4表达与患者5年总生存率关系。Kaplan-Meier法绘制生存曲线,应用单因素及多因素Cox回归分析影响患者5年总生存率的因素。[结果] 结肠癌癌组织及其癌旁组织中PLOD2表达阳性率分别为68.75%(66/96)、25.00%(24/96)(P<0.05),VSIG4表达阳性率分别为56.25%(54/96)、18.75%(18/96)(P<0.05)。Spearman相关性分析显示,结肠癌癌组织中PLOD2与VSIG4表达呈正相关(r=0.363,P=0.041)。PLOD2表达与肿瘤分化程度、浸润深度、淋巴结转移、TNM分期相关(P<0.05);VSIG4表达与肿瘤浸润深度、淋巴结转移、TNM分期相关(P<0.05)。PLOD2阳性表达者5年总生存率为18.03%,阴性表达者为42.67%(χ2=6.969,P=0.008);VSIG4阳性表达者5年总生存率为17.85%,阴性表达者为41.42%(χ2=9.308,P=0.002)。单因素分析结果显示:TNM分期、浸润深度、分化程度、淋巴结转移、PLOD2表达、VSIG4表达是影响患者5年总生存率的因素(P均<0.05);多因素生存分析显示:TNM分期、PLOD2表达是影响患者5年总生存率的独立影响因素(P<0.05)。[结论] 结肠癌患者癌组织中PLOD2、VSIG4表达均升高,两者可作为判断病情及预测预后的因素。

      Abstract:[Objective] To explore the expression and significance of procollagen-lysine 2-xoglutarate 5-dioxygenase 2 (PLOD2) and V-set and immunoglobulin domain-containing 4 (VSIG4) in colon cancer. [Methods] Samples of cancer tissue and adjacent tissues were collected from 96 patients with stage Ⅰ~Ⅳ colon cancer admitted. The expression of PLOD2 and VSIG4 was detected by immunohistochemical method, and their relationship with clinical and pathological parameters of patients was analyzed. The overall 5-year survival(OS) of patients was analyzed with Kaplan-Meier curve and the factors affecting 5-year OS of patients were examined by univariate and multivariate Cox regression analysis. [Results] The positive rates of PLOD2 and VSIG4 expression in colon cancer tissue and its adjacent tissues were 68.75%(66/96) and 25.00%(24/96); 56.25%(54/96) and 18.75%(18/96), respectively(both P<0.05). Spearman correlation analysis showed a positive correlation between PLOD2 and VSIG4 expression in colon cancer tissue(r=0.363, P=0.041). The expression of PLOD2 was correlated with the degree of differentiation, depth of infiltration, lymph node metastasis, and TNM staging(all P<0.05), and the expression of PLOD2 was correlated with the depth of infiltration, lymph node metastasis, and TNM staging(all P<0.05). The 5-year OS of PLOD2 positive expression patients was 18.03%, while negative expression patients were 42.67%(χ2=6.969, P=0.008). The 5-year OS of VSIG4 positive expression patients was 17.85%, while that of negative expression patients was 41.42%(χ2=9.308, P=0.002). Univariate analysis showed that TNM staging, depth of infiltration, degree of differentiation, lymph node metastasis, expression of PLOD2 and VSIG4 were associated with lower 5-year OS in patients(all P<0.05). Multivariate analysis showed that TNM staging and PLOD2 expression were independent impact factors for lower 5-year OS in patients(P<0.05). [Conclusion] The expression of PLOD2 and VSIG4 is elevated in cancer tissues of colon cancer patients, which can serve as biomarkers for predicting prognosis of patients.
