      摘 要:环状RNA(circ RNA)是一类通过反剪接产生的连接3′和5′端的共价键构成的环状非编码RNA(non-coding RNA,ncRNA)分子。结直肠癌是现阶段世界上第三常见的恶性肿瘤,死亡率呈上升趋势。环状RNA因其特殊结构目前已证实在细胞生长发育、肿瘤的发生及进展尤其是结直肠癌中发挥重要作用,有望成为新型的潜在分子标志物。研究证实环状RNA通过充当miRNA“海绵”、RNA结合蛋白、激活表观遗传分子及Wnt/β-catenin通路活性影响结直肠癌增殖、分化、迁移及侵袭,并以此为基础激活EGFR/RAF/MAPK等靶点以及增强结直肠癌细胞敏感性、降低耐药性,从而达到治疗结直肠癌的效果。全文就环状 RNA在结直肠癌中的增殖、分化、迁移、侵袭作用机制以及影响结直肠癌细胞敏感性、耐药性发挥治疗作用等方面进行综述,进一步阐明环状 RNA作为新型生物分子在结直肠癌诊断及治疗中的潜在价值。

      Abstract: Circular RNA (circRNA) is a type of non-coding RNA (ncRNA) covalently bonding at 3′ and 5′ ends by reverse splicing. Colorectal cancer (CRC) is the third most common malignancy in the world with an increasing mortality. CircRNA plays an important role in normal cell growth and progression, as well as tumorigenesis of various malignant tumors including CRC. Studies have demonstrated that circRNA affects CRC proliferation, differentiation, migration and invasion by acting as miRNA “sponge”, targeting RNA-binding protein, activating epigenetic molecules and Wnt/β-catenin pathway; it also regulates EGFR/RAF/MAPK pathway, enhances sensitivity and reduces resistance of CRC cells to chemotherapy. This paper reviews the mechanisms of circRNA in proliferation, differentiation, migration, invasion, drug sensitivity and resistance of CRC; the potential value of circRNA as a novel biomolecule in the diagnosis and treatment of CRC is also discussed.
