      摘 要:[目的] 探索血小板反应蛋白-5(thrombospondin-5,THBS5)在卵巢癌中的表达,并寻找THBS5可能的天然产物抑制剂。[方法] 使用GEPIA数据库评估卵巢癌中5个THBS家族成员的mRNA表达。通过免疫组织化学染色法和Western blot 实验检测THBS5在69例高级别浆液性卵巢癌中表达,采用Western blot 检测THBS5在不同卵巢癌细胞株中的表达,CCK-8 法检测不同药物浓度作用 48 h 后A2780细胞的抑制率。[结果] 40例(57.9%)高级别浆液性卵巢癌组织中检测到THBS5阳性表达,在20例正常卵巢组织中有4例(20.0%)THBS5阳性表达,肿瘤组织中THBS5的表达水平显著高于正常卵巢组织(P<0.05)。一定浓度的小豆蔻明、紫草素、金盏花苷E和姜黄素都能抑制A2780细胞增殖,抑制作用呈浓度效应依赖性关系; 金盏花苷E会导致THBS5蛋白表达的下降。[结论] 高级别浆液性卵巢癌组织中THBS5的表达水平明显高于正常卵巢组织。金盏花苷E能抑制A2780细胞THBS5的表达。

      Abstract:[Objective] To explore the expression of thrombospondin-5(THBS5) in ovarian cancer, and to screen its natural inhibitors. [Methods] GEPIA dataset was used to evaluate the mRNA expression of 5 THBS family members in ovarian cancer. The expression of THBS5 in 69 cases of high-grade serous ovarian cancer(HGSOC) was detected by immunohistochemical staining and Western blot, and the expression of THBS5 was also detected in ovarian cancer cell lines by Western blot. Ovarian cancer cell lines were treated with various drugs at different concentrations and cell proliferation was detected by CCK-8 assay after 48 h. [Results] The immunohistochemistry showed that positive expression of THBS5 was detected in 40 of 69 HGSOC tissue samples(57.9%) and 4 of 20 normal ovarian tissue samples(20.0%). The Western blot also showed that the expression of THBS5 in HGSOC tissues was significantly higher than that in normal ovarian tissues(P<0.05). Cardamonin, shikonin, calunduloside E and curcumin inhibited the proliferation of ovarian cancer A2780 cells in a concentration-dependent manner. Meanwhile, calunduloside E also decreased the expression of THBS5 protein in A2780 cells. [Conclusion] The expression of THBS5 in HGSOC cancer tissues is significantly higher than that in normal ovarian tissues. Calunduloside E can inhibit cell proliferation and the expression of THBS5 in A2780 cells.
