      摘 要:肺癌与各种基因突变、表观遗传修饰改变及染色体改变均有关。反义lncRNA是长链非编码RNA中最多的一类,它在肺癌的发生发展中起着重要作用,从转录前到转录后都有发挥作用。根据其亚细胞定位,可以从不同水平进行调控机制研究,定位于细胞核主要作用是转录前水平和转录水平的调控,位于细胞质则主要是转录后水平的调控。全文阐述反义lncRNA介导靶基因的调控在肺癌中的功能研究,尤其是这些转录本可招募表观遗传调控因子调控靶基因启动子表观遗传修饰状态,介导转录因子与靶基因的结合调控靶基因转录,形成RNA-RNA双链或调节RNA结合蛋白与靶基因的关联来调控靶基因的稳定性,改变靶基因蛋白泛素化修饰调控靶蛋白的表达。

      Abstract: Lung cancer is related with various gene mutations, epigenetic modification and chromosomal alterations. Antisense lncRNA, the most abundant of long non-coding RNA, plays significant roles in the tumorigenesis,the regulating mechanisms of which can be in different levels and subcellular locations: pre-transcriptional and transcriptional in nucleus, and post-transcriptional in cytoplasm. This review is focusing on antisense lncRNA-mediated regulation of relevant gene expression in lung cancer. In particular, these transcripts regulate epigenetic modification state of target genes by recruiting eigenetic regulation factors; regulate interest gene transcription through mediating the binding of transcription factors to target genes; regulate target gene stability by forming RNA-RNA duplex or mediating the related RNA binding protein and interest genes; and regulate the expression of target protein to change its ubiquitination modification.
