      摘 要:[目的] 探讨肌酸激酶线粒体1A(creatine kinase mitochondrial 1A,CKMT1A)在子宫内膜样腺癌(endometrioid adenocarcinoma,ECa)组织和癌旁组织中的表达情况及其对子宫内膜癌细胞Ishikawa增殖和迁移能力的影响。[方法] 选择2017年1月至2020年12月在郑州大学第三附属医院就诊,具有完整临床信息资料的ECa组织及癌旁组织各39例,采用qRT-PCR检测组织中CKMT1A mRNA表达,采用免疫组化SP法检测CKMT1A蛋白表达情况。体外细胞实验使用siRNA干扰Ishikawa细胞中的CKMT1A基因表达,通过CCK8和划痕实验检测敲减CKMT1A后对Ishikawa细胞增殖和迁移能力的影响。[结果] 与癌旁组织相比,ECa组织中CKMT1A mRNA表达量(t=4.006,P<0.05)和CKMT1A的阳性表达率均较高(P<0.001)。CKMT1A mRNA在不同ECa分期的患者组织中表达量不同,差异具有统计学意义(F=11.780,P<0.001)。在敲减CKMT1A组中,细胞增殖(P<0.001)和迁移能力(F=164.400,P<0.001)明显下降。 [结论]CKMT1A在ECa组织中呈高表达,敲减Ishikawa细胞中的CKMT1A后细胞增殖和迁移能力受到抑制。

      Abstract:[Objective] To explore the expression of creatine kinase mitochondrial 1A(CKMT1A) in tissues and adjacent tissues of endometrioid adenocarcinoma and its effect on proliferation and migration of Ishikawa cells. [Methods] A total of 78 cases of endometrioid adenocarcinoma and adjacent tissue with complete clinical information were selected from The Third Affiliated Hospital of Zhengzhou University from January 2017 to December 2020. qRT-PCR and immunohistochemistry assay were used to detect the relative expression of CKMT1A mRNA and the protein expression of CKMT1A. In vitro, siRNA interferencetargeting CKMT1A was applied to down-regulate the expression of CKMT1A in Ishikawa cells. The effect on cell proliferation and migration were detected by CCK8 and scratch-wound assay, respectively. [Results] The expression of CKMT1A mRNA(t=4.006, P<0.05) and protein(P<0.001) in endometrioid adenocarcinoma tissues were higher than those in adjacent tissues. There were significant differences in CKMT1A mRNA expression among patients with different endometrioid adenocarcinoma stages(F=11.780, P<0.001). In CKMT1A knockdown group, cell proliferation(P<0.001) and migration ability(F=164.400, P<0.001) decreased. [Conclusion] CKMT1A was highly expressed in endometrioid adenocarcinoma tissues, and the cell proliferation and migration ability decreased after knockdown of CKMT1A in Ishikawa cells.
