      摘 要:[目的] 探讨异染色质蛋白1结合蛋白3(heterochromatin protein 1 binding protein 3,HP1BP3)在结直肠癌(colorectal cancer,CRC)中的表达及意义。[方法] 通过外源性RNA干扰技术结合细胞增殖、侵袭和迁移体外实验探讨HP1BP3的肿瘤生物学效应。免疫组织化学技术分析313例结直肠癌中HP1BP3的表达,并探讨其表达与患者临床病理特征的相关性。[结果] 外源性敲低HP1BP3表达与SW480细胞增殖、侵袭和转移抑制相关。313例结直肠癌中,HP1BP3高表达104例,低表达209例,HP1BP3表达与患者年龄、性别、肿瘤原发部位及病理分期均无明显相关性(P均<0.05),与淋巴结转移情况密切相关(P=0.001)。[结论]HP1BP3低表达与结直肠肿瘤细胞发生发展抑制相关,其高表达与结直肠癌淋巴结转移相关,提示进展的结直肠癌中可能存在异常的染色质重塑。

      Abstract: [objective] To investigate the expression of heterochromatin protein 1 binding protein 3(HP1BP3) in colorectal cancer(CRC) and its significance. [Methods] The biological effects of HP1BP3 on tumor were investigated by exogenous RNA interference technique combined with cell proliferation, invasion and migration in vitro. The expression of HP1BP3 in 313 cases with colorectal cancer were analyzed by immunohistochemical technique(IHC),and the relationship between the HP1BP3 expression and the clinicopathological features of colorectal cancer was analyzed. [Results] In SW480 cells, non-coding RNA silencing resulted in the decrease of HP1BP3 expression, and the exogenous HP1BP3 knockdown was associated with the inhibition of proliferation, invasion and metastasis. Among the 313 colorectal cancers, 104 cases had high expression of HP1BP3 and 209 cases had low expression. The expression of HP1BP3 was not significantly correlated with the age, gender, primary tumor site and pathological stage of the patients(all P<0.05), but closely correlated with lymph node metastasis(P=0.001). [Conclusion] The low expression of HP1BP3 is associated with the inhibition of colorectal cancer cell development, and its high expression is associated with lymph node metastasis in colorectal cancer, suggesting that abnormal chromatin remodeling be presented in advanced CRC.
