      摘 要:[目的] 分析小细胞肺癌(small cell lung cancer,SCLC)中AXL表达与SCLC临床病理特征的关系。[方法] 利用公共数据库分析SCLC中AXL的表达特征,对与AXL有显著性相关的基因进行GO及KEGG富集分析;应用免疫组化检测SCLC组织中AXL、Vimentin、E-cadherin的表达。[结果] CCLE数据库检索结果显示SCLC细胞系的AXL表达水平最低(P<0.05);Oncomine数据库检索结果显示SCLC组织中AXL的表达明显低于正常肺组织(P<0.05)或与正常肺组织无统计学差异(P>0.05)。CCLE 数据库筛选出 5 255 个在 SCLC 中与 AXL 表达有相关性的基因,其中EMT相关基因 VIM、MMP-2、MMP-9、ZEB-1、ZEB-2 和 TWIST-1与AXL表达呈正相关(P<0.05),N-cadherin和E-cadherin与AXL 表达呈负相关(P<0.05);免疫组化结果显示SCLC中AXL、Vimentin的表达与临床病理参数无相关性,E-cadherin表达与肿瘤分期显著性相关(P=0.001),且AXL与Vimentin、E-cadherin表达不存在线性关系。[结论] AXL在SCLC中表达较低且与EMT相关基因相关,免疫组化结果显示AXL在SCLC组织中的表达水平低于正常肺组织,与Vimentin、E-cadherin的表达无直线影响关系。

      Abstract: [Objective] To analyze the correlation of anexelekto(AXL) expression with clinicopathological features of small cell lung cancer(SCLC) based on bioinformatic study. [Methods] The public database was used to analyze the expression of AXL in SCLC, and the enrichment analysis of AXL-related genes were performed with GO and KEGG databases. The expression of AXL, Vimentin and E-cadherin in 60 SCLC tissue samples was detected by immunohistochemistry. [Results] CCLE database analysis showed that the expression level of AXL in SCLC cell line was the lowest(P<0.05). Oncomine database analysis showed that AXL expression in SCLC tissue was lower than that in normal lung tissue(P<0.05). A total of 5 255 genes related to AXL expression in SCLC were screened from CCLE database, among which VIM, MMP-2, MMP-9, ZEB-1, ZEB-2 and TWIST-1 were positively correlated with AXL expression(P<0.05), while N-cadherin and E-cadherin were negatively correlated with AXL expression(P<0.05). Immunohistochemical results showed that the expression of AXL and Vimentin in SCLC was not correlated with clinicopathological parameters, but the expression of E-cadherin was significantly correlated with tumor stage(P=0.001). AXL expression had no significant linear relationship with Vimentin and E-cadherin expression. [Conclusion] The expression of AXL in SCLC is down-regulated, and the expression level of AXL in SCLC is lower than that in normal lung tissue, and it has no significant linear relationship with the expression of Vimentin and E-cadherin.
