      摘 要:[目的] 探究蒲公英萜醇通过调控α/β水解酶域11反义RNA1(ABHD11-AS1)对膀胱癌细胞侵袭转移的作用,并分析相关机制。[方法] 将对数生长期的人膀胱癌细胞系EJ细胞分为空白对照组(细胞不做特殊处理)、蒲公英萜醇组(给予蒲公英萜醇100 μmol/L处理24 h)、蒲公英萜醇-pcDNA组(转染pcDNA 24 h给予细胞蒲公英萜醇100 μmol/L处理24 h)、蒲公英萜醇-pcDNA-ABHD11-AS1组(转染pcDNA-ABHD11-AS1 24 h后细胞给予蒲公英萜醇100 μmol/L药物处理24 h)。实时荧光定量PCR法检测细胞ABHD11-AS1水平;细胞增殖-毒性8检测试剂盒(CCK-8)法检测细胞增殖;划痕实验、Transwell实验分别检测细胞迁移、侵袭能力;蛋白免疫印迹法检测上皮间质转化标志蛋白E-钙黏附蛋白(E-cadherin)、N-钙黏附蛋白(N-cadherin)、波形蛋白(Vimentin)表达变化。[结果] 与空白对照组比较,蒲公英萜醇组EJ细胞ABHD11-AS1水平、侵袭细胞数、细胞迁移率、N-Cadherin、Vimentin蛋白水平均降低(P<0.05),EJ细胞增殖抑制率、E-cadherin蛋白水平升高(P<0.05)。与蒲公英萜醇组、蒲公英萜醇-pcDNA组比较,蒲公英萜醇-pcDNA-ABHD11-AS1组EJ细胞ABHD11-AS1水平、侵袭细胞数、细胞迁移率、N-Cadherin、Vimentin蛋白水平均升高(P<0.05),EJ细胞增殖抑制率、E-cadherin蛋白水平降低(P<0.05)。[结论] 蒲公英萜醇能够抑制膀胱癌细胞侵袭及迁移,可能是通过抑制ABHD11-AS1介导的上皮间质转化发生实现的。

      Abstract: [Objective] To investigate the effect of dandelion terpenol(DT) on invasion and migration of bladder cancer cells via regulating antisense RNA1 of alpha/beta hydrolase domain 11(ABHD11-AS1). [Methods] Human bladder cancer EJ cells were divided into blank control group, DT group, DT-pcDNA group and DT-pcDNA-ABHD11-AS1 group. DT group was treated with 100 μmol/L DT for 24 h, DT-pcDNA group was transfected with pcDNA, then treated with 100 μmol/L DT for 24 h, and DT-pcDNA-ABHD11-AS1 group was transfected with pcDNA-ABHD11-AS1, then treated with 100 μmol/L DT for 24 h. The levels of ABHD11-AS1 were detected by real-time fluorescent quantitative PCR, cell proliferation was detected by cell counting kit-8(CCK-8), the ability of cell migration and invasion was detected by scratch test and Transwell test, the expression of E-cadherin, N-cadherin and Vimentin was detected by Western blot. [Results] Compared with blank control group, the ABHD11-AS1 level, cell invasion, cell migration, N-Cadherin and Vimentin protein levels in DT group were significantly decreased(P<0.05), the proliferation inhibition rate and E-cadherin protein level increased(P<0.05). Compared with DT group and DT-pcDNA group, the ABHD11-AS1 level, cell invasion, cell migration, N-Cadherin and Vimentin protein levels in DT-pcDNA-ABHD11-AS1 group were increased(P<0.05) , the cell proliferation inhibition rate and E-cadherin protein level decreased(P<0.05). [Conclusion] Dandelion terpenol can inhibit the invasion and migration of bladder cancer cells, which may be associated with inhibition of lncRNA ABHD11-AS1-mediated EMT.
