摘 要: [目的] 分析血浆样本中的差异蛋白在小细胞肺癌(small cell lung cancer,SCLC)诊断及预后中的价值。[方法] 首先应用GSH-CAA-X00蛋白芯片技术检测4例SCLC治疗前后及4名健康人外周血,结合生物信息学分析筛选差异蛋白。应用ELISA检测36例SCLC治疗前后及8名健康人血浆样本中关键差异蛋白胃泌素释放肽前体(progastrin releasing peptide,proGRP)、干扰素诱导T细胞趋化因子(interferon-inducible T cell alphachemoattractant,I-TAC)的表达,分析其表达与SCLC临床指征之间的关系。[结果]蛋白芯片结果显示在健康人和 SCLC 患者治疗前血浆样本中符合筛选且有统计学意义的差异蛋白有 PDGF-AB、S100A8、GHR、GP73、GRO、proGRP、I-TAC等,在 SCLC 患者治疗前后血浆样本中符合筛选且有统计学意义的差异蛋白有 MMP-3、NSE、LRIG3、proGRP、I-TAC等。PDGF-AB、S100A8、GHR、GP73、GRO、MMP-3、NSE、proGRP、I-TAC在健康人、SCLC患者治疗前、治疗后的表达趋势为先上升后下降,其中proGRP、I-TAC在两组间的变化最为显著且都具有统计学意义。ELISA结果显示proGRP、I-TAC与芯片结果一致,即与健康人相比,在SCLC患者表达升高,经治疗后下降,但与临床指征无相关性。[结论] 差异蛋白尤其是proGRP、I-TAC可能是SCLC的潜在诊治肿瘤标志物,单独使用或与其他常用生物标志物联合使用可能有助于SCLC的早期诊断及预后判断。 |

Abstract:[Objective] To investigate plasma proteins with potential value for diagnosis and prognosis of small cell lung cancer(SCLC). [Methods] The differentially expressed proteins were detected with GSH-CAA-X00 protein chip and analyzed with bioinformatics from plasma samples of 4 cases of SCLC before and after treatment, and 4 healthy subjects. The key differential proteins progastrin releasing peptide(proGRP) and interferon-inducible T cell alpha chemoattractant(I-TAC)were detected with ELISA in plasma samples of 36 SCLC patients before and after treatment and 8 healthy subjects. [Results] The proteins differentially expressed between SCLC patients before treatment and healthy subjects were PDGF-AB, S100A8,GHR,GP73,GRO,proGRP,I-TAC,etc. The proteins differentially expressed in SCLC patients before and after treatment were MMP-3,NSE,LRIG3,I-TAC,etc. Compared with healthy controls, the plasma levels of PDGF-AB,S100A8,GHR,GP73,GRO,MMP-3,NSE,LRIG3,proGRP,I-TAC in SCLC patients were significantly increased, then decreased after treatment, among which the changes of proGRP and I-TAC were the most significant. ELISA results showed that the changes of proGRP and I-TAC were consistent with the chip results, however, which were not correlated with clinical indications. [Conclusion] Differentially expressed proteins in plasma, especially proGRP and I-TAC, may be used as potential tumor markers for diagnosis and prognosis of SCLC. |
