| 摘 要：随着致病机制的深入研究，发现一种新型的跨膜蛋白Claudin18(CLDN18)具有高选择性表达的特点。在生理状态下，CLDN18只表达在肺和正常胃黏膜，而在人类消化系统肿瘤中，CLDN18的异常表达导致生长因子的异常流入，通过刺激上皮细胞自分泌和旁分泌增加，导致上皮内瘤变和浸润性肿瘤的发生。目前CLDN18.2特异性靶向药物claudiximab（IMAB362）已在多项临床试验显示，其单独治疗或与化疗相联合都具有良好的疗效和安全性。全文就CLDN18在消化系统肿瘤中的表达作用以及药物研发现状进行分析。 |

| Abstract： Claudin18(CLDN18) is a novel transmemmembrane protein with highly selective expression in human tissues. Under physiological conditions， CLDN18 is only expressed in lung and normal gastric mucosa， while in gastrointestinal tumors， the upregulated expression of CLDN18 leads to abnormal inflow of growth factors， which increase the autocrine and paracrine of epithelial cells， thus leading to intraepithelial neoplasia and invasive tumors. The recently developed CLDN18.2?鄄targeted drug Claudiximab(IMAB362) has shown good efficacy and safety in a number of clinical trials for cancer treatment as used alone or combined with chemotherapy. In this article， the expression of CLDN18 in digestive tract tumors and the status quo of related drug development are reviewed. |
