      摘 要:[目的] 探讨肿瘤坏死因子-α(TNF-α)通过影响核因子-κB(NF-κB)信号通路对宣威肺癌恶性生物学行为的调控。[方法] ELISA检测临床收集宣威肺癌、非宣威肺癌及正常人血清样本中TNF-α水平。RT-qPCR、Western blot检测人支气管上皮样细胞16HBE、人肺腺癌细胞A549及宣威肺癌细胞XWLC-05中TNF-α;CCK-8检测细胞增殖水平;流式细胞术检测细胞凋亡率及细胞周期;Transwell检测细胞迁移能力;Western blot检测NF-κB经典及非经典信号通路相关蛋白水平。[结果] TNF-α在宣威肺癌患者血清及细胞中显著性高表达,差异均有统计学意义(P<0.05)。沉默TNF-α不仅阻滞细胞周期、抑制细胞增殖及迁移,并促进凋亡(P<0.05);也抑制NF-κB经典信号通路。XWLC-05被外源性TNF-α促进的恶性生物学行为可被NF-κB信号通路抑制剂扭转(P<0.05)。[结论] TNF-α通过激活NF-κB经典信号通路促进宣威肺癌细胞恶性生物学行为。

      Abstract: [Objective] To investigate the regulation of tumor necrosis factor-α(TNF-α) on malignant biological behavior of Xuanwei lung cancer by influencing nuclear factor-κB(NF-κB) signal pathway. [Methods] Serum samples of Xuanwei lung cancer, lung cancer and normal people were collected, then the level of TNF-α was detected by ELISA. RT-qPCR and Western blot was used to detect the expression of TNF-α mRNA and protein in normal human bronchial epithelioid cell line 16HBE, human lung adenocarcinoma cell line A549 and Xuanwei lung cancer cell line XWLC-05. Cell proliferation, apoptosis rate, cell cycle and migration were measured by CCK-8, flow cytometry and transwell assay. Moreover, the expression of protein of classical and noncanonical NF-κB signal pathway related proteins were detected by Western blot. [Results] TNF-α was highly expressed in serum and cell line of Xuanwei lung cancer(P<0.05), silencing TNF-α significantly blocked the cell cycle, inhibited cell proliferation and migration, and promoted cell apoptosis(P<0.05). In addition, silencing TNF-α markedly inhibited the classical NF-κB signal pathway. Further experiments confirmed that NF-κB signal pathway inhibitor reversed the promotion of malignant biological behavior of Xuanwei lung cancer which was caused by TNF-α(P<0.05). [Conclusion] TNF-α promotes the malignant biological behavior of Xuanwei lung cancer cells by activating the classical NF-κB signal pathway.
