摘 要:叉头框 (forkhead box,FOX)是一类具有进化上高度保守的翼-螺旋 (Winged?鄄helix)DNA结合域的转录因子超家族,在肿瘤发生发展中发挥重要调节作用。FOXK1/2是近年研究的热点,既可通过调节肿瘤代谢、自噬、增殖、侵袭转移促进肿瘤的发生发展,亦可通过转录抑制肿瘤相关基因表达发挥抑癌作用,还可以影响肿瘤治疗应答。FOXK1/2作用机制的进一步阐明,对于肿瘤发生机制的揭示、肿瘤预后预测及肿瘤新治疗靶点的发现均具有重要意义。 |

Abstract:Forkhead box proteins comprise a transcription factor superfamily containing an evolutionally conserved Winged-helix DNA binding domain,and play an important role in tumorigenesis and progression,of which FOXK1/2 have become the research hotspots in recent years. Growing evidence shows that FOXK1/2 can not only promote the development of cancer by regulating tumor metabolism,autophagy,proliferation,invasion and metastasis,but also exert tumor suppression roles by transcriptionally regulating the expression of tumor associated genes,and also affect the response of anti-tumor treatment. Further elucidating the regulatory mechanisms of FOXK1/2 will be of great significance in tumor pathogenesis,prognostic prediction and discovery of novel therapeutic targets. |
