摘 要:程序性细胞死亡因子4 (programmed cell death 4,Pdcd4)作为抑癌基因,具有抑制肿瘤细胞生长、侵袭和转移等功能,在各类肿瘤中表达下调。虽然肿瘤细胞中尚未发现Pdcd4基因突变,但生长因子和白细胞介素等微环境因素可以调节其基因的表达。目前认为Pdcd4的作用方式一方面可以与部分转录因子结合从而调节其下游基因的表达,另一方面可以与翻译起始因子eIF4A结合抑制其解螺旋酶的活性从而抑制蛋白质的翻译,但Pdcd4如何通过翻译抑制来调节肿瘤发生的机制仍不清楚。全文着重阐述了Pdcd4在肿瘤中的表达及其调控因素,以及其发挥抑制肿瘤作用可能依赖的机制,为抗肿瘤研究提供新的思路。 |

Abstract:As a tumor suppressor gene,programmed cell death 4(Pdcd4) has the functions of inhibiting growth,invasion and metastasis of tumor cells,which expression was downregulated in various types of tumors. Although Pdcd4 gene mutations have not been found in tumor cells,microenvironmental factors such as growth factors and interleukins can regulate their gene expression. Up to now,it is believed that Pdcd4 can not only bind to some transcription factors to regulate the expression of its downstream genes,but also can inhibit the translation of proteins through binding to the translation initiation factor eIF4A which helicase activity is essential for translation. However,the mechanism of Pdcd4 playing roles in tumorigenesis and tumor development remains unclear. In this review,we discuss the regulation of Pdcd4 expression and the mechanism of Pdcd4 playing roles in tumor suppression,which may provide new ideas for anti-cancer research. |
