      摘 要:[目的]探讨lncRNA-KAT7在晚期鼻咽癌患者中的表达及对顺铂化疗敏感性的影响。[方法] 选取86例晚期鼻咽癌患者的组织标本作为研究组,选取同时间段就诊鼻咽良性病变患者的组织标本86例作为对照组。比较鼻咽癌组织、癌旁组织和鼻咽良性组织lncRNA-KAT7的相对表达量;研究组患者诱导化疗后评价疗效,根据疗效分为敏感组和抵抗组,比较lncRNA-KAT7表达水平。比较人鼻咽癌细胞株CNE1、HONE1、C666-1、CNE-2、5-8F及鼻咽上皮细胞株NP69中lncRNA-KAT7表达水平。细胞计数试剂盒(CCK)检测转染KAT7-siRNA过表达载体对细胞增殖和凋亡的影响。[结果] 鼻咽癌组织lncRNA-KAT7相对表达量为1.05±0.31,低于癌旁组织(1.47±0.48)和鼻咽良性组织(2.59±0.73),差异有统计学意义(P<0.05)。化疗敏感组lncRNA-KAT7相对表达量为1.23±0.34,高于抵抗组(0.78±0.21),差异有统计学意义(P<0.05)。NP69细胞中lncRNA-KAT7相对表达量为1.01±0.11,高于CNE1(0.16±0.03)、HONE1(0.31±0.07)、C666-1(0.23±0.04)、CNE-2(0.32±0.20)、5-8F(0.39±0.16)细胞,差异均有统计学意义(P均<0.05)。KAT7-siRNA组lncRNA-KAT7相对表达量为5.36±1.15,高于阴性对照组(1.27±0.12)和空白对照组(1.24±0.13),差异有统计学意义(P<0.05)。KAT7-siRNA组的IC50为12.54±3.43,低于阴性对照组(18.23±3.25),差异有统计学意义(P<0.05)。在顺铂1、4、8、16及32μmol/L浓度下KAT7-siRNA组细胞凋亡率明显高于阴性对照组,差异有统计学意义(P<0.05)。[结论] lncRNA-KAT7在鼻咽癌组织中低表达,上调lncRNA-KAT7可以提高鼻咽癌对顺铂的敏感性。

      Abstract:[Objective] To investigate the expression of lncRNA-KAT7 in patients with nasopharyngeal carcinoma(NPC) and its effect on chemosensitivity. [Methods] The relative expression levels of lncRNA-KAT7 was detected with RT-PCR in cancer tissues and adjacent tissues of 86 NPC patients(NPC group),and 86 samples of benign nasopharyngeal tissues(control group). The efficacy after induction chemotherapy was evaluated,and patients were divided into a sensitive group and a resistance group according to the efficacy. The expression levels of lncRNA-KAT7 in human nasopharyngeal carcinoma cell lines CNE1,HONE1,C666-1,CNE-2,5-8F and nasopharyngeal epithelial cell line NP69 were compared. Cell counting kit(CCK) was used to detect the proliferation and apoptosis after NPC cells were transfected with KAT7-siRNA. [Results] The relative expression of lncRNA-KAT7 in nasopharyngeal carcinoma was 1.05±0.31,which was lower than that in adjacent tissues(1.47±0.48) and benign nasopharyngeal tissue(2.59±0.73)(P<0.05). The relative expression level of lncRNA-KAT7 in the sensitive group was(1.23±0.34),which was higher than that in the resistance group(0.78±0.21)(P<0.05). The relative expression of lncRNA-KAT7 in NP69 cells was(1.01±0.11),which was higher than that of NPC cell lines CNE1(0.16±0.03),HONE1(0.31±0.07),C666-1(0.23±0.04),CNE-2(0.32±0.20) and 5-8F(0.39±0.16)(all P<0.05). The relative expression level of lncRNA-KAT7 in the KAT7-siRNA group was(5.36±1.15),which was higher than that in the negative control group(1.27±0.12) and the blank control group(1.24±0.13)(P<0.05). The IC50 of the KAT7-siRNA group was(12.54±3.43),which was significantly lower than that of the negative control group(18.23±3.25)(P<0.05). The apoptosis rate of the KAT7-siRNA group was significantly higher than that of the negative control group at the concentrations of cisplatin 1,4,8,16,and 32 μmol/L(P<0.05). [Conclusion] lncRNA-KAT7 is under-expressed in nasopharyngeal carcinoma,and up-regulation of lncRNA-KAT7 can increase the sensitivity of nasopharyngeal carcinoma to cisplatin.
