      摘 要:[目的] 探讨结肠肿瘤组织中紧密连接蛋白Claudin-1的表达与血清炎症因子的相关性,以及它们与患者临床病理特征、预后的关系。[方法] 选择结肠癌患者57例,采用RT-PCR和Western Blot方法检测肿瘤组织和癌旁组织中Claudin-1的表达,采用ELISA的方法检测患者血清炎症因子水平,分析两者的相关性以及两者与患者临床病理特征的关系。绘制3年生存曲线,分析Claudin-1和炎症因子与患者预后的关系。[结果] Claudin-1 mRNA和蛋白均在癌旁组织中低表达,在肿瘤组织中高表达(1.34±0.36 vs 3.24±1.47,P=0.005;0.92±0.28 vs 2.41±1.25,P=0.007);肿瘤患者血清TNF-α、IL-6、IL-1β水平均明显高于健康人群(P=0.010、0.003、0.001);Spearman检验发现Claudin-1表达和血清炎症因子水平呈正相关(P<0.05);Claudin-1蛋白表达及血清炎症因子水平均与TNM分期、分化程度、淋巴结转移有关(P<0.05)。Log-rank (Mantel-Cox)检验发现Claudin-1表达和血清炎症因子水平均是影响患者预后的因素。 [结论] 结肠肿瘤组织中Claudin-1的表达和血清炎症因子存在相关性,两者均是患者预后的影响因素。

      Abstract:[Objective] To explore the relationship between the expression of claudin-1 in colon tumor tissues and serum inflammatory cytokines,and their relationship with the clinicopathological characteristics and prognosis of patients.[Methods] The mRNA and protein expression of Claudin-1 in tumor tissues and pericancerous tissues of 57 colon cancer patients were detected by RT-PCR and Western blot,respectively. The serum levels of inflammatory cytokines of patients were measured by ELISA. The relationship between expression of claudin-1 and serum inflammatory cytokines with clinicopathological characteristics and the prognosis was analyzed. [Results] The expression of claudin-1 mRNA and protein in cancer tissues was significantly higher than that in pericancerous tissues(3.24±1.47 vs. 1.34±0.36,P=0.005;2.41±1.25 vs. 0.92±0.28,P=0.007). Serum levels of TNF-α,IL-6,IL-1β of patients were significantly higher than those of healthy subjects(P=0.010,0.003 and 0.001). Spearman test revealed a positive correlation between claudin-1 expression and serum inflammatory cytokines(P<0.05). Chi-square analysis found that both the levels of claudin-1 and inflammatory cytokines were related to TNM stage,degree of differentiation and lymph node metastasis(all P<0.05). Log-rank(Mantel-Cox) test showed that both the levels of claudin-1 expression and inflammatory cytokines were associated with the prognosis of patients. [Conclusion] The expression of claudin-1 in colon tumor tissues is correlated with serum inflammatory cytokines,and both of them are factors affecting the prognosis of patients.
