      摘 要:[目的] 探讨了Lnc SNHG16通过miR-140-5p/wnt1对结直肠癌细胞增殖和迁移的影响。[方法] 选取结直肠癌肿瘤组织和癌旁组织各102例,采用荧光定量PCR检测Lnc SNHG16和miR-140-5p表达水平和两者间的关系。采用生物信息学和双荧光素酶报告基因分析Lnc SNHG16和miR-140-5p之间的关系以及靶基因;在结肠癌细胞系SW480建立Lnc SNHG16敲降和miR-140-5p过表达细胞系,采用Western blot分析Lnc SNHG16和miR-140-5p对目的蛋白的影响;采用CCK8分析不同处理结肠癌细胞增殖;采用Transwell分析不同处理的结肠癌细胞的迁移。[结果] 与癌旁组织相比,结直肠癌组织中Lnc SNHG16表达水平显著增加,而miR-140-5p表达水平显著下调,差异均有统计学意义(P<0.05)。生物信息学研究发现Lnc SNHG16调控着miR-140-5p的表达水平,并调控wnt1蛋白的表达,调节WNT/β-Catenin信号通路活性。Lnc SNHG16敲降或过表达miR-140-5p可显著抑制SW480细胞的增殖和迁移,差异均有统计学意义(P<0.05)。[结论] Lnc SNHG16通过靶向作用于miR-140-5p/wnt1轴进而调节着结直肠癌细胞的增殖和迁移。

      Abstract:[Objective] To investigate the effects of Lnc SNHG16 on proliferation and migration of colorectal cancer cells and its mechanism. [Methods] The expression levels of Lnc SNHG16 and microRNA-140-5p in cancer tissues and adjacent colorectal tissues from 102 patients with colorectal cancer were detected by fluorescence quantitative PCR. Bioinformatics and double luciferase reporter gene were used to analyze the relationship between Lnc SNHG16 and microRNA-140-5p and target genes. Lnc SNHG16 knockdown and microRNA-140-5p overexpression cell lines were established from colon cancer cell line SW480. The effects of Lnc SNHG16 and microRNA-140-5p on target proteins were analyzed by Western blot. CCK8 was used to analyze the proliferation of colon cancer cells with different treatments,and Transwell was used to analyze the migration of colon cancer cells with different treatments. [Results] Compared with the adjacent colorectal tissues,the expression of Lnc SNHG16 was significantly increased in colorectal cancer tissues,while the expression of miR-140-5p was significantly decreased(P<0.05). Bioinformatics studies found that Lnc SNHG16 regulated the expression of microRNA-140-5p,the expression of Wnt1 protein and the activity of WNT/beta-Catenin signaling pathway. Down-regulation or over-expression of microRNA-140-5p in Lnc SNHG16 significantly inhibited the proliferation and migration of SW480 cells(P<0.05). [Conclusion] Lnc SNHG16 regulates the proliferation and migration of glioma cells by targeting the microRNA-140-5p/wnt1 axis.
