      摘 要:[目的]测定直肠癌患者癌组织中LOH12CR1 mRNA和蛋白的表达水平并分析其临床意义。[方法] 取72例直肠癌患者的直肠癌组织及癌旁正常组织,应用实时聚合酶联法和Western Blot法检测直肠癌和癌旁正常组织LOH12CR1 mRNA和蛋白的表达水平,分析直肠癌组织LOH12CR1 mRNA和蛋白表达水平与患者临床病理特征和预后的相关性。[结果] (1)直肠癌患者癌组织LOH12CR1 mRNA和蛋白的表达水平均较癌旁正常组织显著下调(P<0.05);(2)LOH12CR1 mRNA和蛋白表达水平与患者年龄、性别、肿瘤直径、距肛缘距离无相关性(P>0.05),但与肿瘤分化程度、TNM分期、Dukes分期和淋巴结转移具有相关性(P<0.05);(3)术后6个月时出现肿瘤复发/转移的患者其术前LOH12CR1 mRNA和蛋白较未复发/转移患者显著降低(P<0.05);(4)Cox回归分析结果显示,LOH12CR1 mRNA和蛋白水平、肿瘤分化程度、TNM分期、Dukes分期、淋巴转移是影响直肠癌患者临床预后的独立因素(P<0.05)。[结论] 抑癌基因LOH12CR1可能参与直肠癌的发生、发展,并与患者术后临床预后有关。

      Abstract:[Objective] To investigate the expression and clinical significance of LOH12CR1mRNA and protein in rectal cancer. [Methods] The rectal cancer tissue and normal pericancerous tissue were taken from 72 patients with rectal cancer. The mRNA and protein expression of LOH12CR1 in rectal cancer tissue and pericancerous tissue were determined by real-time polymerase chain reaction(qRT-PCR) and Western blotting,and the relationship of mRNA and protein expression level of LOH12CR1 in rectal cancer tissue with clinicopathological features and clinical prognosis was analyzed. [Results] The mRNA and protein expression levels of LOH12CR1 in rectal cancer tissues were significantly down-regulated compared with those in pericancerous tissue(P<0.05). The mRNA and protein expression levels of LOH12CR1 were not correlated with age,gender,tumor diameter,and distance from anal edge(P>0.05),but were correlated with tumor differentiation,TNM stage,Dukes stage and lymphatic metastasis(P<0.05). The pre-operative mRNA and protein levels of LOH12CR1 in patients with tumor recurrence/metastasis 6 months after operation were significantly decreased compared with those in patients without recurrence/metastasis(P<0.05). Cox regression analysis showed that mRNA and protein levels of LOH12CR1,differentiation,TNM stage,Dukes stage and lymphatic metastasis were independent prognostic factors of patients with rectal cancer(P<0.05). [Conclusion] Tumor inhibitory gene LOH12CR1 may be involved in the occurrence and development of rectal cancer and is related to the postoperative clinical prognosis of patients.
