摘 要:核糖体结合蛋白1 (RRBP1)参与调控未折叠蛋白反应(UPR),该反应几乎涉及到癌症发展的各方面。近年研究表明,RRBP1在多种恶性肿瘤中高表达(与免疫组化评估的正常组织相比),是内质网应激(ERS)和调控未折叠蛋白反应(UPR)相关成员。RRBP1通过增强葡萄糖调节蛋白78(GRP78)表达减轻内质网应激,减少细胞凋亡,帮助癌细胞生存。在应激条件下,La蛋白介导RRBP1的翻译,与RRBP1内部核糖体进入位点(IRES)相结合并调节其活性,是体内RRBP1 IRES的激活剂。全文主要针对RRBP1与ERS、UPR、GRP78之间调控关系、La蛋白介导的翻译及其在相关肿瘤发生发展中的作用进行综述。 |

Abstract:Ribosome-binding protein 1(RRBP1) participates in the regulation of unfolded protein response(UPR),which is involved in almost all aspects of cancer development. Previous studies have shown that compared with normal tissues,RRBP1 is highly expressed in multiple malignancies as assessed by immunohistochemistry(IHC). RRBP1 is a member of the ER stress response(ERS) and associated unfolded protein response(UPR). Ectopic RRBP1 overexpression can alleviate intracellular stress-induced apoptosis and protect tumor survival through the enhancement of GRP78. La autoantigen(La) induces RRBP1 expression through internal ribosome entry site(IRES)-mediated translation during cellular stress condition and is identified as an activator of RRBP1 IRES in vivo. In this review,the regulatory roles of RRBP1 in protecting tumor cell from ERS,adjusting UPR,enhancing the expression of GRP78,and La-mediated translation in relation to tumorigenesis of associated cancers are summarized. |
