      摘 要:[目的] 探究GOLM1对宫颈癌上皮间质转化的影响及其与临床病理之间的关系。[方法]选取76例宫颈鳞状细胞癌手术标本和50例正常宫颈组织标本作为研究对象,采用免疫组化法检测GOLM1在宫颈癌组织及正常宫颈组织中的表达水平,并分析GOLM1蛋白表达与宫颈癌组织临床病理特征之间的关系。构建GOLM1正常表达及GOLM1低表达Hela细胞系,Western blot检测GOLM1蛋白与E-cadherin、Vimentin表达水平变化;采用Transwell实验检测GOLM1蛋白对宫颈癌Hela细胞侵袭能力的影响。[结果] 与正常宫颈组织中GOLM1蛋白的阳性表达率(22.0%)相比,GOLM1蛋白在宫颈癌组织中的阳性表达率(72.3%)升高(P<0.05)。GOLM1蛋白在不同年龄组、肿瘤直径中的表达无显著性差异(P>0.05);但在不同FIGO分期、分化程度、淋巴结转移以及深肌层浸润的宫颈癌组织中的表达则有显著性差异(P<0.05)。与对照组细胞相比,shRNA组细胞GOLM1和Vimentin蛋白表达水平显著性下调,E-cadherin表达水平则显著性升高(P<0.05)。与对照组侵袭细胞数(465.72±40.19)相比,shRNA组侵袭细胞数(103.36±9.89)显著性降低(P<0.05)。[结论] GOLM1表达与宫颈癌的侵袭转移存在相关性,为宫颈癌的诊断和治疗提供了新的思路。

      Abstract:[Objective] To investigate the effect of GOLM1 expression on epithelial-mesenchymal transition and its relationship with clinicopathological features of cervical cancer. [Methods] The expression of GOLM1 was detected by immunohistochemistrin in 76 specimens of cervical squamous cell carcinoma tissues and 50 specimens of normal cervical tissues. The relationship between the GOLM1 expression and clinicopathological features of cervical cancer was analyzed. The GOLM1 normal expression Hela cells(control group) and GOLM1 knockdown Hela cells(shTNA group) were constructed. The expression levels of GOLM1,E-cadherin and vimentin in Hela cells were detection by Western blot,and the cell invasive ability was analyzed by Transwell assay. [Results] Compared with the normal cervical tissues,the expression of GOLM1 protein in cervical cancer tissues was significantly increased(72.3% vs. 22%,P<0.05). The expression of GOLM1 was not significantly associated with the age and tumor diameter,while it was significantly associated with FIGO stages,tumor differentiation,lymph node metastasis and deep muscle infiltration in cervical cancer tissues(P<0.05). Compared with control Hela cells,the expression of GOLM1 protein and vimentin in shRNA-transfected Hela cells was significantly down-regulated,while the expression of E-cadherin was significantly increased(P<0.05). The number of invading cells in shRNA group was significantly lower than control group(103.36±9.89 vs. 465.72±40.19,P<0.05). [Conclusion] There is a significant correlation between the expression of GOLM1 and the invasion of cervical cancer,indicating that GOLM1 might be used as a biomarker for cervical cancer.
