摘 要:[目的] 观察辐射旁效应在细胞间的再传递。[方法] 用6MV-X射线照射鼠卵巢癌NUTU19细胞株,其培养液制备一代条件培养液,处理一代效应细胞鼠源NUTU19细胞株、肠黏膜上皮IEC-6细胞株,由一代鼠源效应细胞的分泌物为二代条件培养液,再处理二代效应细胞NUTU19细胞株、IEC-6细胞株和鼠淋巴细胞,检测细胞生长活力和凋亡水平。[结果]NUTU19二代培养液处理细胞48h后,对IEC-6和NUTU19细胞的凋亡率有促进提高作用(P<0.05),对淋巴细胞的凋亡率有降低作用(P<0.05)。NUTU19二代培养液处理细胞72h后,促进NUTU19、IEC-6的凋亡(P<0.05),对淋巴细胞凋亡率无影响(P>0.05)。IEC-6二代培养液处理细胞48h后,对NUTU19、IEC-6的凋亡有促进作用(P<0.05),对淋巴细胞的凋亡有降低作用(P<0.05)。IEC-6二代培养液处理细胞72h后,对IEC-6、淋巴细胞的凋亡均无影响(P>0.05)。对NUTU19的凋亡有促进作用(P<0.05)。[结论] 在一定的条件下,肿瘤细胞、肠上皮细胞、淋巴细胞都能够再传递辐射旁效应的损伤。 |

Abstract:[Objective] To observe the retransmission of radiation side effects between cells. [Methods] The mouse ovarian cancer cell line NUTU19 was irradiated with 6MV-X-rays,and the culture medium was prepared for the first-generation conditioned medium. The first-generation effector cells were used to detect the NUTU19 cell line and the intestinal mucosal epithelial IEC-6 cell line. The secretion of effector cells was a second-generation conditioned medium,and the second-generation effector cells NUTU19,IEC-6 and mouse lymphocytes were treated to measure cell viability and apoptosis.[Results] After treated with NUTU19 second-generation medium for 48 hours,the apoptosis rate of IEC-6 and NUTU19 cells was promoted(P<0.05),and the apoptosis rate of lymphocytes was decreased(P<0.05). After treatment with NUTU19 second-generation medium for 72h,the apoptosis of NUTU19 and IEC-6 was promoted(P<0.05),and there was no effect on the apoptosis rate of lymphocytes(P>0.05). After treatment with IEC-6 second-generation medium for 48h,the apoptosis of NUTU19 and IEC-6 was promoted(P<0.05),and the apoptosis of lymphocytes was decreased(P<0.05). After treated with IEC-6 second-generation medium for 72h,there was no effect on IEC-6 and lymphocyte apoptosis(P>0.05). It promoted the apoptosis of NUTU19(P<0.05). [Conclusion] Under certain conditions,tumor cells,intestinal epithelial cells and lymphocytes can retransmit the damage of the side effect of radiation. |
