      摘 要:[目的] 探讨microRNA-184(miR-184)在人肾组织细胞病变以及癌症发生发展中的生理功能。[方法] 采用反转录-荧光实时定量PCR(RT-qPCR)检测肾癌组织细胞、癌旁组织细胞中miR-184相对表达;在体外进行细胞实验,向786-0肾癌细胞系转染 miR-184 mimics,并运用MTT噻唑兰法对转染miR-184 mimics成功的肾癌细胞进行体外增殖活力检测,同时通过流式细胞仪来检测细胞凋亡发生的情况;Western-blot法检测目的蛋白的相对表达水平。[结果] 荧光定量PCR检测结果表明,miR-184在肾癌组织中的表达水平显著低于癌旁组织(P<0.001);细胞体外转染实验结果表明,与阴性对照组相比,转染miR-184 mimics的786-0肾癌细胞株,其细胞增殖活力显著下降而凋亡率显著升高,差异均有统计学意义(P<0.05);Western-blot检测结果表明,在miR-184 mimics的786-0肾癌细胞株中,EPB41L5表达水平显著下降,相比于阴性对照组以及空白对照组差异均有统计学意义(P<0.05)。[结论]肾癌患者的癌组织中miR-184的表达显著降低,其可能参与肾癌的发生发展;体外细胞学实验表明其在肾癌细胞的增殖和凋亡过程中起着重要作用,可能是通过调控细胞内源EPB41L5蛋白的表达来发挥其生理功能。

      Abstract:[Objective] To explore the expression of microRNA-184(miR-184) in human renal cell carcinoma and its effect on cell proliferation and apoptosis. [Methods] The miR-184 expression in cancer tissue and adjacent renal tissue samples from 49 patients with renal cell carcinoma was detected by RT-qPCR. Mimics of miT-184 were transfected into human renal carcinoma 786-0 cells,and the proliferation of the transfected cells was detected by MTT thiazolam assay,and the cell apoptosis was detected by flow cytometry. The relative expression level of target protein was detected by Western-blot. [Results]The result of quantitative PCR showed that the expression of miRNA-184 in renal cell carcinoma was significantly lower than that in adjacent tissues(P<0.001). Compared with the negative control group,the cell proliferation of 786-0 cells transfected with miR-184 mimics was significantly decreased and the apoptosis rate was significantly increased(P<0.05). Western-blot showed that in miR-184 mimics-transfected 786-0 cells,the expression level of EPB41L5 was lower than that of negative control group and the blank control group(P<0.05).[Conclusion] The expression of miR-184 decreases significantly in renal cell carcinoma,and in vitro study shows that it may inhibit proliferation and enhance apoptosis of renal cell carcinoma cells through regulating the expression of EPB41L5 protein.
