摘 要:[目的] 检测IL-17与趋化因子在胃癌组织中的表达,探讨IL-17在胃癌组织中聚集及其促胃癌进展的作用机制。[方法]收集35例手术切除胃癌组织标本,包括癌组织和非癌对照组织,采用荧光定量RT-PCR法检测IL-17及趋化因子CCL2、CCL7、CCL17、CCL20、CCL22、CXCL2和CXCL5 mRNA的表达;分析IL-17与表达增高的趋化因子的相关性;采用免疫组织化学染色法,检测胃癌组织内CD34和CD66b的表达。[结果] 与非癌对照组织相比,胃癌组织中IL-17和趋化因子CCL2、CCL7、CCL20、CCL22 mRNA表达显著增高(P<0.05),而趋化因子CCL17、CXCL2和CXCL5 mRNA表达在两组间无明显差异(P>0.05)。胃癌组织中IL-17 mRNA表达水平升高与趋化因子CCL20、CCL22 mRNA表达水平升高明显相关(r=0.7901,P=0.0038;r=0.9683,P<0.0001),而与趋化因子CCL2、CCL7 mRNA表达水平升高无明显相关性(P>0.05)。IL-17 mRNA高表达组胃癌组织内CD34和CD66b的表达水平明显高于IL-17 mRNA低表达组(P<0.05)。[结论] 胃癌组织内IL-17表达增高可能与胃癌组织高表达细胞因子CCL20、CCL22,趋化Th17细胞由外周循环向肿瘤募集有关,增高的IL-17通过促进新生血管生成和中性粒细胞浸润促进肿瘤进展。 |

Abstract:[Objective] To investigate the expression of IL-17 and related chemokines in gastric cancer tissues and their relation to cancer progression. [Methods] Tissue samples of gastric cancer and normal gastric mucosa were collected from 30 patients with gastric cancer. The mRNA expression of IL-17,and chemokines CCL2,CCL7,CCL17,CCL20,CCL22,CXCL2,CXCL5 were detected by qRT-PCR. The correlation between expression IL-17 and chemokines was determined by liner regression analysis. The expression of CD34 and CD66b in gastric cancer tissues were examined by immunohistochemical staining. [Results] Compared to normal mucosa,the mRNA expressions of IL-17,CCL2,CCL7,CCL20 and CCL22 in cancer tissues were significantly up-regulated(P<0.05),while no difference was observed in mRNA expression of CCL17,CXCL2 and CXCL5(P>0.05). The high expression of IL-17 mRNA in tumor tissues were positively correlated with the expression of CCL20(r=0.7901,P=0.0038)and CCL22(r=0.9683,P<0.001),but not correlated with CCL2 and CCL7(P>0.05). Furthermore,the expressions of CD34 and CD66b in gastric cancer with high expression of IL-17 mRNA were significantly higher than those with low expression of IL-17mRNA.[Conclusion] The up-regulated expression of IL-17 mRNA in tumor tissues is associated with overexpression of CCL20 and CCL22 mRNA,indicating that IL-17 may promote tumor progression by inducing neovascularization and neutrophil infiltration. |
