      摘 要:[目的] 探讨STAT3与miR-21的调控关系以及其对舌鳞癌细胞Tscca对顺铂(DDP)敏感性的影响作用。[方法] 体外培养Tscca细胞和Tscca/DDPR细胞,将细胞分为空白对照组、阴性对照组、WP1066处理组、DDP处理组和WP1066+DDP共处理组。采用MTT法和划痕实验检测DDP和/或WP1066对Tscca细胞和Tscca/DDPR细胞增殖和迁移的影响;采用RT-PCR法和Western blot法检测不同处理方式Tscca细胞和Tscca/DDPR细胞miR-21、STAT3、p-STAT3的表达水平。[结果] 经不同浓度DDP(0.25、1、4、16μg/ml)处理后,Tscca细胞的增殖和迁移能力明显低于Tscca/DDPR细胞(P<0.05),且具有剂量依赖性。Tscca细胞中miR-21、STAT3表达量明显低于Tscca/DDPR细胞(P<0.05)。WP1066可以明显抑制Tscca细胞和Tscca/DDPR细胞的增殖和迁移活性,且两组细胞比较差异无统计学意义(P>0.05)。经WP1066处理后,Tscca细胞、Tscca/DDPR细胞miR-21和p-STAT3表达水平均低于空白对照组(P<0.05)。而DDP对Tscca/DDPR细胞miR-21和p-STAT3表达水平无明显影响,与空白对照组比较差异无统计学意义(P>0.05)。Pearson相关性分析显示,miR-21与p-STAT3的表达量呈正相关(r=0.999,P<0.05)。[结论] STAT3/miR-21过表达与舌鳞癌细胞对顺铂的耐药性相关,有可能成为舌鳞癌患者对化疗药物产生耐药性的潜在干预靶点。

      Abstract:[Objective] To investigate the relationship of signal transducer and activators of transcription 3(STAT3) and miR-21 expression and the sensitivity to cisplatin(DDP) in tongue squamous cell carcinoma. [Methods] Tongue squamous cell carcinoma Tscca cells and DDP-resistance Tscca cells(Tscca/DDPR) were cultured in vitro and divided into control group,negative control group,WP1066 treatment group,DDP treatment group and WP1066+DDP treatment group. The proliferation and metastasis ability of Tscca cells and Tscca/DDPR cells were detected by MTT and scratch healing assay,respectively. The mRNA and protein expressions of miR-21,p-STAT3,STAT3 in Tscca cells and Tscca/DDPR were detected by RT-PCR and Western blot,respectively. [Results] The proliferation and metastasis ability of Tscca cells treated with DDP(0.25,1,4,16μg/ml) were lower than those of Tscca/DDPR cells(P<0.05) in a dose-independent manner. The miR-21,STAT3 expressions in Tscca cells were lower than those in Tscca/DDPR cells(P<0.05). There was no difference in the proliferation and metastasis between Tscca and Tscca/DDPR cells treated with WP1066(P>0.05). And the miR-21 and p-STAT3 expressions in Tscca and Tscca/DDPR cells treated with WP1066 were lower than those in control group and negative control group(P<0.05). There was no difference in miR-21 and p-STAT3 expressions in Tscca/DDPR cells treated with DDP compared with control group(P>0.05). The miR-21 expression was positively correlated with p-STAT3(r=0.999,P<0.05). [Conclusion] The down-regulation of miR-21/STAT3 may enhance the sensitivity of tongue squamous cell carcinoma to DDP.
