      摘 要:[目的] 探讨盐酸川芎嗪联合顺铂对小鼠Lewis肺癌细胞侵袭黏附能力的影响及其作用机制。[方法] 小鼠Lewis肺癌细胞体外培养,细胞黏附侵袭实验检测细胞侵袭能力的改变,Western Blot检测各组细胞Twist、E-cadherin、N-cadherin蛋白表达的变化。[结果] 经DDP与TMP干预36h后,与对照组相比,用药组细胞的侵袭能力呈剂量依赖性下降,且DDP联合TMP组与单独使用DDP组相比,抑制细胞侵袭的能力更为显著(P<0.05,P<0.01)。细胞黏附实验显示,与对照组(23.76±1.64)相比,单独应用顺铂(47.40±0.85)可显著性增高细胞黏附抑制率(P<0.01),而单独应用TMP需药物浓度达到2000μg/ml时才可显著性增高黏附抑制率(P<0.01),两药联合组黏附抑制率均高于单独使用DDP组,尤以高剂量组抑制率最为显著(P<0.01)。与单独用药相比,DDP+TMP可降低Twist、N-cadherin蛋白,并上调E-cadherin蛋白的表达(P<0.01)。[结论] 盐酸川芎嗪联合顺铂能抑制小鼠Lewis肺癌细胞的侵袭和转移,其机制可能与直接抑制肿瘤细胞侵袭,抑制相关基因蛋白Twist及N-cadherin,促进黏附分子E-cadherin蛋白表达有关。

      Abstract:[Objective] To investigate the effect of tetramethylpyrazine(TMP) and cisplatin(DDP) on migration and invasion ability of mouse Lewis lung cancer(LLC). [Methods] The cultured LLC cells were treated with DDP,TMP and DDP+TMP,respectively. The cell invasion ability was determined with cell adhesion assay,the expressions of Twist,E-cadherin and N-cadherin in LLC cells were detected by Western bloting. [Results] After 36h of treatment,compared with the control group,the invasion and migration of LLC cells in DDP,TMP and DDP+TMP groups were inhibited in a dose-dependent manner; and the effect of DDP+TMP was significantly stronger than that of TMP and DDP alone(P<0.05,P<0.01). The adhesion assay showed that compared with control group(23.76±1.64),the inhibition rate of adhesion in DDP group(47.40±0.85)and high dose-TMP group(65.727.78)was dramatically increased(P<0.01),and that in DDP+TMP group,especially in DDP+ high dose-TMP group was significantly higher than that in DDP group(P<0.01). Western blot showed that TMP and DDP reduced the expression of Twist,N-cadherin and increase the expression of E-cadheri(P<0.01). [Conclusion] Tetramethylpyrazine and DDP can inhibit invasion and metastasis of LLC cells,which may be associated with inhibition of Twist,N-cadherin gene expression and increase of the adhesion molecule E-cadherin expression.
