| 摘 要：［目的］ 探讨重楼皂甙Ⅰ对胃癌肿瘤相关成纤维细胞（cancer-associated fibroblasts，CAFs）的作用。［方法］ 以体外原代培养的胃癌CAFs为研究对象，MTT法检测重楼皂甙Ⅰ对CAFs细胞的增殖抑制作用，Western Blot法检测CAFs细胞FAP、SPARC、SDF-1、TNC和HGF蛋白表达改变。［结果］ 不同浓度重楼皂甙Ⅰ能有效抑制CAFs细胞的增殖，且呈浓度依赖性。2μg/ml重楼皂甙Ⅰ作用CAFs细胞24h后，FAP、SDF-1和HGF蛋白表达降低，与对照组相比差异均有统计学意义（P＜0.05）。SPARC和TNC蛋白表达改变与对照组相比差异无统计学意义（P＞0.05）。［结论］ 重楼皂甙Ⅰ能抑制胃癌CAFs细胞的体外增殖，通过下调FAP、SDF-1和HGF蛋白表达影响CAFs功能。 |

| Abstract：［Objective］ To explore the effect of polyphyllin Ⅰ（PPⅠ） on gastric cancer-associated fibroblasts. ［Methods］ The primary gastric CAFs were obtained by tissue culture. The effects of PPⅠ on cell proliferation were detected by MTT method. The expressions of FAP，SPARC，SDF-1，TNC and HGF were detected by Western Blot. ［Results］ The growth of CAFs cells was inhibited by PPⅠ in dose-dependent manner. Compared with control groups，the expressions of FAP，SDF-1 and HGF protein decreased significantly（P＜0.05）after treated with 2μg/ml PPⅠ at 24h，while the expressions of SPARC and TNC protein were not statistically significant（P＞0.05）. ［Conclusion］ PPⅠ can inhibit the cell proliferation of gastric CAFs and affect the function of gastric CAFs by down-regulating the expression of FAP，SDF-1 and HGF protein. |
