摘 要:[目的] 探讨缺氧诱导因子-2α(HIF-2α)与securin在乳腺癌组织中的表达及其对无瘤生存率的意义。[方法] 采用免疫组化SP法检测70例乳腺癌组织中HIF-2α和securin的表达,并分析其与乳腺癌临床疾病特征的关系及相关性。对患者进行无瘤生存期的随访,Kaplan Meier生存曲线分析HIF-2α和securin对无瘤生存期的意义。[结果] 70例乳腺癌组织中 HIF-2α和securin阳性率、高表达率分别为30.0%(21/70)和34.3%(24/70)。Spearman相关分析表明,HIF-2α与securin的表达呈正相关(r=0.381,P=0.001)。Securin高表达和HIF-2α阳性与乳腺癌患者绝经情况、雌激素受体、孕酮受体、Her-2、淋巴结转移、ki67、TNM分期和肿瘤大小均无关(P>0.05)。Kaplan Meier生存曲线表明,HIF-2α阳性患者的无瘤生存期明显高于阴性患者。Cox模型多因素生存分析结果显示,HIF-2α是乳腺癌无瘤生存期的独立影响因素(P<0.05)。[结论] HIF-2α和securin与乳腺癌的发生、发展密切相关,尤其是HIF-2α可能成为判断乳腺癌术后无瘤生存率的独立有效指标。 |

Abstract:[Objective] To investigate the expression of HIF-2α and securin in breast cancer and its relation to disease-free survival. [Methods] The expression of HIF-2α and securin was detected by SP immunohistochemistry in 70 patients with breast cancer. The disease-free survival(DFS) of patients was examined by Kaplan-Meier survival curve and the relationship of HIF-2α and securin expression with clinical features and DFS was analyzed. [Results] The expressions of HIF-2α and securin were 30.0%(21/70)and 34.3%(24/70)in breast cancer,respectively. Spearman correlation analysis revealed that HIF-2α and securin were significantly correlated with each other(r=0.381,P=0.001). The expression of HIF-2α or securin was not correlated with menopausal status,PR,ER,HER-2,lymphatic metastasis,Ki67,TNM and size of tumor(all P>0.05). Kaplan-Meier survival curve showed the DFS of patients with HIF-2α positive expression was higher than those with negative expression. Cox regression model analysis revealed that HIF-2α was an independent prognostic factor of disease free survival(P<0.05). [Conclusion] The expression of HIF-2α was closely correlated with securin in breast cancer. HIF-2α may be an independent protective factor of disease-free survival in postoperative patients with breast cancer. |
