      摘 要:[目的] 分析血浆miR-143表达与食管鳞癌浸润肌层深度及淋巴结转移的关系,为食管鳞癌转移的早期防治及作用机制提供理论学依据。[方法] 收集食管鳞癌患者术前一周血标本40例,同期收集40例健康体检者血标本,运用实时定量荧光PCR检测两组人群血浆中miR-143表达水平,探讨食管鳞癌患者血浆中miR-143表达水平与食管鳞癌深肌层浸润及淋巴结转移的关系。[结果] 与健康人群相比,食管鳞癌患者血浆中miR-143表达水平降低,差异有统计学意义(P<0.001)。食管鳞癌患者血浆miR-143表达水平与年龄(P=0.154)、性别(P=0.131)、肿瘤部位(P=0.092)均无关;T3+T4期患者血浆中miR-143表达水平低于T1+T2期患者(P=0.008);与无深肌层浸润患者相比,有深肌层浸润患者血浆miR-143表达水平降低(P=0.012);有淋巴结转移的患者血浆中miR-143表达水平较无淋巴结转移患者低(P=0.003)。[结论] 食管鳞癌患者血浆miR-143表达水平较健康人群下调,且在有深肌层浸润和发生淋巴结转移的食管鳞癌患者血浆中表达水平明显下调,其有望作为评估食管鳞癌患者肿瘤进展的新生物学标志。

      Abstract:[Objective] To investigate the relationship between miR-143 expression and esophageal squamous cell carcinoma(ESCC),and to provide a theoretical basis for the early prevention and treatment of ESCC metastasis. [Methods] Forty blood samples from patients with esophageal squamous cell carcinoma and 40 samples from healthy subjects were collected. The expression of miR-143 in plasma was detected by real-time quantitative PCR. The relationship between plasma levels of miR-143 and ESCC in deep ESCC and lymph node metastasis were analyzed.[Results] Compared with the healthy population,the expression of miR-143 in ESCC was significantly decreased(P<0.001). The expression level of miR-143 in the plasma of patients with ESCC was not correlated with age(P=0.154),gender(P=0.131),or tumor location(P=0.092). Patients at T3 + T4 stage had a lower expression level of miR-143 than those at T1+T2 stage(P=0.008),and patients with myometrial invasion and lymph node metastasis had a lower expression level of miR-143 than those without(P=0.012 and 0.003,respectively). [Conclusion] The expression of miR-143 in ESCC is lower than that in healthy people,and is down-regulated in patients with deep myometrial invasion and lymph node metastasis. It may be used as a new biological marker for assessing tumor progression in ESCC.
