| 摘 要：［目的］ 探讨脑胶质瘤标本中Mipu1和HIF-1ɑ表达及其相关性。［方法］ 胶质瘤患者24例，根据WHO病理分级分低级别组12例（Ⅰ级5例，Ⅱ级7例），高级别组12例（Ⅲ级7例，Ⅳ级5例），以其中10例低级别胶质瘤患者瘤旁部分脑组织作为对照组。对三组标本进行针对Mipu1和HIF-1的免疫组化、实时荧光定量核酸扩增检测和免疫印迹分析检测，对所得得数据进行统计分析。［结果］ 免疫组化结果显示Mipu1和HIF-1ɑ表达蛋白均主要定位在细胞核内，且有随着胶质瘤病理级别升高而增强表达的趋势。三组标本的Mipu1和HIF-1ɑ的相对mRNA表达量和相对蛋白表达量组间差异有统计学意义（P<0.05）；且Mipu1和HIF-1ɑ 在胶质瘤中的表达呈正相关（P<0.05）。［结论］ HIF-1ɑ和Mipu1在胶质瘤中表达升高，且两者表达呈正相关。 |

| Abstract：［Objective］ To investigate the expression and their correlation of HIF-1α and Mipu1 in human glioma.［Methods］ 24 patients with glioma were classified as low grade group of 12 cases(5 cases of grade Ⅰ，7 cases of grade Ⅱ) and the high grade group of 12 cases(7 cases of grade Ⅲ，5 cases of grade Ⅳ) according to theWHO pathological grading. Tumor adjacent tissues form 10 patients with low grade glioma were selected as the control group. The expression of Mipu1 and HIF-1 of the three groups were detected by immunohistochemical staining，real-time quantitative PCR and Western blot，and the data obtained were statistically analyzed.［Results］ The results of immunohistochemical staining showed that the expression of Mipu1 and HIF-1α protein was mainly localized in nucleus，and there was a tendencyofincreasedexpression of Mipu1 and HIF-1α with the increase of pathological grade. The relative mRNA expression quantity and relative protein expression quantityof Mipu1 and HIF-1α was statistically different among the 3 groups(P<0.05). The Mipu1 and HIF-1αexpressionin glioma was positively correlated(P<0.05).［Conclusions］ HIF-1α and Mipu1 are overexpressed in human glioma，and the expression of HIF-1α and Mipu1 showed a positive correlation. |
