      摘 要:[目的] 研究葫芦素B对卵巢癌细胞SKOV3的生长抑制作用。[方法] 利用MTT法检测不同浓度葫芦素B对卵巢癌细胞的增殖抑制作用;流式细胞术检测细胞周期和凋亡情况;荧光定量PCR法测定STAT3 mRNA表达水平。[结果] 葫芦素B对SKOV3细胞的增殖具有抑制作用,并呈现时间和剂量依赖性(P<0.05);随葫芦素浓度的增加,出现明显的G2/M期细胞阻滞,细胞凋亡率也随之增加(P<0.05);STAT3 mRNA表达量明显下降(P<0.05)。[结论] 葫芦素B抑制STAT3的表达,引起细胞周期阻滞和诱导细胞凋亡,抑制卵巢癌细胞的生长。

      Abstract:[Objective] To investigate the inhibitory effect of cucurbitacin B on the growth of human ovarian cancer cells. [Methods] Human ovarian cancer SKOV3 cells were treated with cucurbitacin B of different concentrations in vitro. MTT assay was used to assay the growth of ovarian cancer cells. Flow cytometry was performed to analyze cell cycle and apoptosis rate. Fluorescent quantitative PCR was used to detect the expression of STAT3mRNA. [Results] Cucurbitacin B inhibited SKOV3 cell proliferation in a dose and time dependent manner(P<0.05). With the increasing of cucurbitacin B concentration,SKOV3 cells were arrested at the G2/M phase,the cell apoptosis increased(P<0.05),and the expression of STAT3 mRNA decreased (P<0.05). [Conclusion] Cucurbitacin B can inhibit STAT3 mRNA expression,arrest cell cycle and induce apoptosis of human ovarian cancer SKOV3 cells.
