      摘 要:母体胚胎亮氨酸拉链激酶(MELK)是一种Ser/Thr蛋白激酶,也是一种细胞周期依赖性蛋白激酶,属于蔗糖非发酵-1/AMP活化蛋白激酶(Snf1/AMPK)家族。最近研究发现,MELK在许多肿瘤细胞中表达水平增高,其高表达能够抑制肿瘤细胞及某些干细胞的分化与凋亡,并促进肿瘤细胞增殖,与许多恶性肿瘤的不良预后密切相关,被认为是一种原癌基因。此外,MELK参与多种信号通路调控,促进细胞增殖和肿瘤形成。MELK沉默有望成为肿瘤新的治疗靶点之一。本文就MELK的结构、功能、表达调控及与胃肠道肿瘤关系的研究进展作一综述。

      Abstract:Maternal embryonic leucine zipper kinase (MELK),a member of the sucrose-non-fermenting (SNF1)/AMPK family of serine-threonine kinases,is a cell cycle dependent protein kinase. Recently,overexpression of MELK has been detected in a variety of human tumors,which can inhibit cell differentiation,apoptosis of tumor cells and tumor stem cells,and promote proliferation of tumor cells. MELK expression is correlated with poor prognosis of a variety of cancers,and known as an oncogen. In addition,MELK is involved in the regulation of many signaling pathways,which drive cell proliferation and tumor formation. Knockdown of MELK may be a new target for cancer treatment. This article aims to review the latest research progress of the structure,function and expression regulation of MELK in gastrointestinal cancer.
