摘 要:[目的] 检测LMO1、Bcl-2、Bax、Survivin、Caspase-3蛋白在胃癌组织中的表达,并对这些蛋白之间的关系和临床意义进行分析。 [方法] 对96例胃癌手术患者的肿瘤组织标本及50例癌旁组织标本进行免疫组化染色(SP法),检测LMO1、Bcl-2、Bax、Survivin、Caspase-3蛋白的表达情况。同时收集患者的病理学情况,对与各指标的关系进行分析。 [结果] 胃癌组织中LMO1、Bcl-2、Survivin表达均比癌旁组织增强(均P<0.05),Caspase-3蛋白在胃癌组织中表达低于癌旁组织(P<0.05),Bax在两种组织中表达无明显差异(P>0.05)。胃癌组织中细胞凋亡率明显低于癌旁组织(P<0.01)。胃癌组织中LOM1蛋白强表达者细胞凋亡率明显低于弱表达者(P<0.01)。Spearman相关分析发现,胃癌组织中LMO1与Bcl-2表达存在正相关(r=0.3058,P=0.002),LMO1与Caspase-3表达存在负相关(r=-0.2886,P=0.004);Bcl-2与Survivin表达存在正相关关系(r=0.2986,P=0.0315),Bcl-2与Bax表达呈负相关(r=-0.4062,P<0.001),Survivin与Caspase-3也呈负相关(r=-0.3142,P=0.002)。[结论] 胃癌组织中LMO1蛋白可能通过调节Bcl-2、Caspase-3的表达和活性而参与肿瘤凋亡。 |

Abstract:[Objective] To investigate the relationship between LMO1 expression and cell apoptosis in gastric cancer. [Methods] The expressions of LMO1 and apoptosis-related proteins Bcl-2,Bax,Survivin,Caspase-3 were detected immunohistochemically in 96 gastric cancer specimens and 50 adjacent tissue specimens. The relationship of LMO1 expression with cell apoptosis was analyzed. [Results] Expressions of LMO1,Bcl-2,Survivin proteins in gastric cancer tissues were stronger and expression of Caspase-3 protein was weaker than those in adjacent cancer tissues (all P<0.05); no significant difference in Bax expression was found between gastric cancer tissues and adjacent cancer tissues(P>0.05). Apoptosis rate in gastric cancer tissues was lower than that in adjacent cancer tissues(P<0.01). In gastric cancer tissues,apoptosis rate in cancer with positive expression of LOM1 was lower than that with negative expression of LOM1(P<0.01).Spearman correlation analysis showed that positive correlation was found between LMO1 and Bcl-2(r=0.3058,P=0.002);negative correlation was found between LMO1 and Caspase-3(r=-0.2886,P=0.004);positive correlation was found between Bcl-2 and Survivin(r=0.3385,P=0.0141);negative correlation was found between Bcl-2 and Bax(r=-0.4062,P<0.001),Survivin and Caspase-3(r=-0.3142,P=0.002). [Conclusion] LMO1 protein might be involved in apoptosis of gastric cancer by regulating expression and activity of Bcl-2,caspase-3 proteins. |
