      摘 要:[目的] 观察不同剂量贝伐单抗对肿瘤局部炎症因子表达的影响。[方法] 接种人结肠癌DLD-1细胞的裸鼠24只,随机分为4组。对照组采用0.9%生理盐水,试验1组2.5mg/kg贝伐单抗,试验2组采用5mg/kg贝伐单抗,试验3组采用10mg/kg贝伐单抗,均分别于第1、5、9d注射。第10d处死裸鼠,留取皮下肿瘤。观察4组间肿瘤大小及肿瘤组织内炎症因子IL-1β、TNF-α、PGE-23 mRNA表达的差异。[结果] 对照组及3个试验组间肿瘤体积比较差异无统计学意义。试验3组肿瘤组织内IL-1β mRNA表达的抑制最明显,与其他3组比较均有统计学差异(P=0.022、0.043、0.022);试验3组肿瘤组织内TNF-α mRNA表达的抑制最明显,与对照组、试验2组比较差异有统计学意义(P=0.046、0.004);PGE-23 mRNA表达与贝伐单抗的使用无相关性。[结论] 10mg/kg贝伐单抗治疗后肿瘤组织内炎症指标IL-1β、TNF-α mRNA的表达明显下降,提示贝伐单抗能抑制肿瘤局部炎症反应。

      Abstract:[Purpose] To observe the effect of different doses of bevacizumab on the expressions of local inflammatory factors in tumors. [Methods] DLD-1 human colon cancer cells were inoculated in 24 nude mice,and these mice were randomly divided into four groups:control group,experimental group 1,experimental group 2 and experimental group 3. The mice in these groups were treated with 0.9% normal saline,2.5mg/kg of bevacizumab,5mg/kg of bevacizumab and 10mg/kg of bevacizumab on day 1,5 and 9,respectively. The mice were sacrificed on day 10,and the subcutaneous xenografts were obtained. The differences in tumor sizes as well as IL-1β,TNF-α and PGE-23 mRNA expressions in tumors were investigated. [Results] No significant difference in tumor size was observed between the control group and the experimental groups. The IL-1β mRNA expression in experimental group 3 was significantly lower than that in the other three groups (control group,experimental group 1 and experimental group 2;P=0.022,0.043 and 0.022,respectively). The TNF-α mRNA expression in experimental group 3 was obviously inhibited,and its expression was statistically lower than that in the control group and experimental group 2(P=0.046,0.004). PGE-23 mRNA expression was not affected by bevacizumab treatment. [Conclusion] 10 mg/kg bevacizumab might significantly decrease IL-1β and TNF-α mRNA expressions in tumor tissue,which suggest that bevacizumab can inhibit local inflammatory reactions caused by tumors.
