摘 要:[目的] 研究HPV16 E6和Hippo途径效应分子YAP的相关性。[方法] 用Western Blot、RT-PCR和激光共聚焦显微镜观察HPV16 E6过表达或敲减后YAP的表达和定位,以及其直接作用的下游基因结缔组织生长因子(CTGF)的表达。[结果] HPV16 E6敲减或过表达时YAP相应地表达减少或增加,磷酸化的YAP增加或减少,YAP主要定位于胞浆或胞浆和胞核中,YAP下游CTGF的表达相对应减少或增加。[结论] HPV16 E6诱导Hippo途径失常,YAP的表达升高,可能提升HPV16 E6的致癌性,Hippo途径可能是治疗高危性HPV感染相关疾病的候选药物作用靶标。 |

Abstract:[Purpose] To explore the association between HPV16 E6 and Yap. [Methods] The expression and localization of YAP and its downstream connective tissue growth factor (CTGF) were analyzed using Western Blot,RT-PCR and laser scanning confocal microscope after HPV16 E6 overexpression and knockdown.[Results] HPV16 E6 knockdown or overexpression induced the decreasing or increasing YAP expression,and Yap mainly located in cytoplasm or cytoplasm and nuclear,and CTGF expression decreased or increased.[Conclusion] HPV16 E6 induces Hippo pathway deregulation and Yap overexpression might promote HPV16 E6 pathogenesis,implying that Hippo pathway might be candidate drug target of hr-HPV infection-induced diseases. |
