      摘 要:[目的] 探讨细丝蛋白A(FLNA)在肝癌组织中的表达及过表达对肝癌细胞生物表型的影响。[方法] 采用免疫组织化学方法、Western Blot检测75例肝癌组织及距其癌组织边缘2cm以上的镜下未见癌浸润的37例正常肝脏组织中FLNA的表达情况。采用慢病毒转染建立FLNA过量表达的肝癌HepG2细胞株。荧光定量PCR及Western Blot检测FLNA转染后肝癌HepG2细胞株中FLNA表达含量的变化。MTT法、Transwell检测FLNA过量表达对肝癌细胞增殖及细胞侵袭转移的影响。[结果] 免疫组织化学结果表明,FLNA蛋白在肝癌组织及正常肝脏组织中的表达阳性率分别为37.3%、89.2%,差异有统计学意义(P<0.05)。Western Blot结果表明,FLNA 蛋白在肝癌组织的相对表达量较正常肝脏组织明显降低,两者比较差异具有统计学意义(P<0.05)。FLNA蛋白表达水平与肝癌有无淋巴结转移、临床分期以及组织学分级有关(P<0.05),而与患者年龄、性别、肝癌病理类型及T分期无关(P>0.05)。FLNA高表达的肝癌细胞其增殖能力明显减弱、侵袭转移能力降低,MMP-9蛋白的表达量明显下调。[结论] 肝癌组织中FLNA蛋白表达明显下降,可能是正常肝脏组织恶性转变的重要生物学标志,对预测肝癌发生、浸润转移有重要意义。

      Abstract:[Purpose] To investigate the expression of filamin A (FLNA) in hepatocellular carcinoma and the biological effect on its cell line by FLNA overexpression. [Methods] Immunohistochemistry and Western Blot were used to analyze FLNA protein expression in 75 cases of hepatocellular cancer and 37 cases of normal tissues. FLNA lentiviral vector and empty vector were respectively transfected into hepatocellular cancer HepG2 cell line. Quantitative real-time RT-PCR (qRT-PCR) and Western Blot were used to detect the mRNA level and protein of FLNA. MTT assay,migration and invasion assays were also conducted to detect the influence of the up-regulated expression of FLNA that might be found on HepG2 cell biological effect. [Results] Immunohistochemistry:the level of FLNA protein expression was significantly lower in hepatocellular cancer tissues than that in normal tissues(P<0.05). Western Blot:the relative amount of FLNA protein in hepatocellular cancer tissues was significantly lower than that in normal tissues (P<0.05). The level of FLNA protein expression did not correlate with gender,age,T stage and pathological type(P>0.05),but it correlated with lymph node metastasis,clinic stage and histological grade(P<0.05). The result of biological function showed that HepG2 cell transfected FLNA had a lower survival fraction,decreasing in migration and invasion,and lower Matrix metallopeptidase 9(MMP-9) protein expression compared with HepG2 cell untransfected FLNA (P<0.05). [Conclusion] FLNA expression decreases in hepatocellular cancer and correlates significantly with lymph node metastasis,clinic stage,histological grade,suggesting that FLNA may play important roles as a negative regulator in hepatocellular cancer HepG2 cell by promoting degradation of MMP-9.
