      摘 要:[目的] 通过比较维吾尔族宫颈癌与宫颈非病变组织中Bcl-2、Bcl-xl、c-IAP1表达的差异,了解维吾尔族宫颈癌与凋亡基因的关系及意义。[方法] 采用含20 000条Oligo DNA的人类全基因组寡核苷酸芯片,对5例维吾尔族宫颈癌与宫颈非病变组织成组检测差异表达基因,通过MAS分析得到两组间凋亡相关的差异表达基因:Bcl-2、Bcl-xl、c-IAP1,并采用RT-PCR及免疫组化验证芯片检测其mRNA和蛋白的表达。[结果] 与宫颈非病变组织相比,维吾尔族宫颈癌患者中凋亡相关基因Bcl-2、Bcl-xl和c-IAP1 mRNA和蛋白表达均上调。[结论] 基因芯片筛选出的肿瘤差异基因,对于研究新疆宫颈癌发生的分子机制及宫颈癌的临床早期诊疗具有重要意义,有利于宫颈癌的易感性预测、早期诊断及疾病监测。维吾尔族宫颈癌的发生与细胞增殖、凋亡的动态平衡密切相关,IAP家族和Bcl-2家族抗凋亡基因在维吾尔族宫颈癌组织中高表达可能在宫颈癌抗凋亡机制中起着重要作用。

      Abstract:[Purpose] By comparing genome-wide expression in cervical cancer and normal cervix,to clarify the molecular tumorigenesis mechanism of Bcl-2,Bcl-xl and c-IAP1.[Methods] Gene-expression profiles of 5 samples of cervical cancer and 5 samples of normal cervix were analyzed using oligo microarray containing 20 000 genes.The differentially expressed genes between the two groups were analyzed using the Significance Analysis of Microarrays(MAS) in order to get the specifically differentially expressed apoptosis related genes:Bcl-2,Bcl-xl and c-IAP1. The apoptosis gene expression presence was verified by real-time PCR(RT-PCR) and immunohistochemistry(IHC).[Results] Compared to normal cervix,3 genes of apoptosis signaling pathway,including Bcl-2,Bcl-xl and c-IAP1 up-regulated in cervical cancer.[Conclusions] Proving the occurrence of invasive cervical cancer is a multistep genetic pathways regulated more complex process of biological molecular marker spectrum of cervical cancer and screening of tumor marker gene,to study the molecular mechanism of cervical cancer occured in Xinjiang and the early clinical diagnosis and treatment of cervical cancer has important significance.It is helpful for us to get further understanding on the molecular mechanism of development for study of common differentially expressed genes between them.High expression of IAP family and Bcl2 family anti-apoptotic proteins in cervical cancer tissues may play an important role in cervical cancer anti-apoptotic mechanisms,and these genes can be used for cervical cancer susceptibility prediction not only early diagnosis but also disease monitoring.
