      摘 要:[目的] 评价重楼皂甙Ⅰ对胰腺癌PANC-1细胞增殖和凋亡的影响。[方法] 以体外培养的胰腺癌PANC-1细胞为研究对象,MTT 法检测重楼皂甙Ⅰ对PANC-1细胞增殖的抑制作用,Annexin-Ⅴ-FITC/PI 双染法检测细胞凋亡,Western Blot 法检测PI3K、Akt、pAkt、Bax、Bcl-2、caspase-3蛋白表达改变。[结果] 不同浓度重楼皂甙Ⅰ能有效抑制PANC-1细胞的增殖,且呈时间浓度依赖性(P<0.01)。低、中、高浓度重楼皂甙Ⅰ作用PANC-1细胞24、48h 后,细胞凋亡率明显增加,与对照组相比,具有统计学差异(P<0.01)。2μg/ml 重楼皂甙Ⅰ作用PANC-1细胞48h 后,PI3K、pAkt、Bcl-2蛋白表达降低,caspase-3、Bax蛋白表达增加,与对照组相比,具有统计学差异(P<0.01)。[结论] 重楼皂甙Ⅰ能抑制胰腺癌PANC-1细胞的体外增殖,诱导细胞凋亡,机制可能与降低PI3K、pAkt、Bcl-2 蛋白表达,增加Bax 及caspase-3 蛋白表达有关。

      Abstract:[Purpose] To investigate the effect of Paris Saponin Ⅰ on proliferation and apoptosis in pancreatic carcinoma cell line PANC-1.[Methods] The effects of Paris Saponin Ⅰ on cell proliferation and cell apoptosis were detected by MTT method and Annexin-Ⅴ-FITC/PI respectively. The expressions of PI3K,Akt,pAkt,Bax,Bcl-2 and caspase-3 were detected by Western Blot.[Results] The growth of PANC-1 cells was inhibited by Paris Saponin Ⅰ in dose-dependent and time-dependent manner. Cell apoptosis was induced significantly after treated with different concentration of Paris Saponin Ⅰ at 24h and 48h(P<0.01). The expression of PI3K,pAkt and Bcl-2 protein decreased,while the expressions of Bax and caspase-3 protein increased after treated with 2μg/ml Paris SaponinⅠ at 48h.[Conclusion] Paris Saponin Ⅰ can inhibit cell proliferation of pancreatic carcinoma cell line PANC-1 in dose-dependent and time-dependent manner and induce cell apoptosis,associating with decreasing expression of PI3K,pAkt and Bcl-2,increasing expression of Bax and caspase-3.
