      摘 要:[目的]研究内源性血管生成抑制因子vasobihin-1对人卵巢癌SKOV-3细胞体外增殖能力的影响。[方法] 体外培养SKOV-3细胞,用vasohibin-1重组腺病毒感染SKOV-3细胞,采用蛋白质印迹法检测SKOV-3细胞中vasohibin-1蛋白的表达水平,应用CCK-8法检测不同时间点vasohibin-1重组腺病毒对SKOV-3细胞增殖的抑制能力。[结果] 感染48h后,在SKOV-3细胞中vasohibin-1蛋白表达量与对照组相比显著性增加(P<0.05);25MOI vasohibin-1重组腺病毒感染SKOV-3细胞24h时,过表达组与阴性对照相比,两者之间没有统计学差异(P=0.065);余各组间相比差异均具有统计学意义(P<0.05),且抑制能力随MOI增大而增加(P<0.05),随时间的延长而增加(P<0.05)。[结论] Vasohibin-1过表达能够抑制SKOV-3细胞的增殖能力,并具有浓度和时间依赖性,可为卵巢癌的治疗提供新思路。

      Abstract: [Purpose] To investigate the effects of the endogenous angiogenesis inhibitor vasohibin-1 on proliferation of the human ovarian cancer SKOV-3 cells.[Methods] SKOV-3 cells were cultured in vitro,then transfected by vasohibin-1 recombinant adenovirus. The expression of vasohibin-1 protein in SKOV-3 cells was confirmed by Western blot. The inhibition of proliferation in SKOV-3 cells were estimated by CCK8 in different times.[Results]Compared with control group,the expression of vasohibin-1 protein increased significantly after transfection for 48h(P<0.05).There was no statistical difference between overexpression group and negative control group after 25MOI vasohibin-1 recombinant adenovirus transfected SKOV-3 cells for 24h(P=0.065);but the other groups had statistical significance and the inhibition of vasohibin-1 recombinant adenovirus on the proliferation of SKOV-3 cells increased with the growth of MOI and with the time prolonging(P<0.05). [Conclusion] Overexpression of vasohibin-1 could inhibit the proliferation of SKOV-3 cells,with dose- and time- dependent manners,which may provide a new idea for ovarian cancer treatment.
