      摘 要:[目的] 观察汉黄芩素对人肝癌细胞HepG2细胞株基因表达谱的影响,探索汉黄芩素的抗肿瘤机制。[方法] 用不同浓度的汉黄芩素处理HepG2细胞。应用Affymetrix HG-U133Plus2.0全人类基因表达谱芯片,检测汉黄芩素作用于肝癌细胞48h后的基因表达差异。[结果] 肝癌细胞经汉黄芩素作用48h后,筛出差异表达2倍以上的基因共406个,其中295个基因上调,111个基因下调。这些差异基因主要涉及148功能分类和61个基因通路,包括细胞凋亡、肿瘤发生、代谢、信号转导、细胞周期、物质运输等多个生物过程。[结论] 汉黄芩素抑制肝癌细胞生长与其对GADD45α、CDKN1A、CDKN2B、JUN和EGFR等基因的表达调控有关。

      Abstract: [Purpose] To study the effects of wogonin on gene expression of human hepatocellular cancer cell line HepG2 and elucidate the anticancer mechanisms of Wogonin. [Methods] HepG2 cell line was treated in the presence of different concentrations of wogonin. Gene chip was applied to evaluate the gene expression profile of hepatocellular cancer cells by wogonin treated 48h. [Results] A total of 406 differentially expressed genes were observed.295 genes up-regulated,and 111 genes down-regulated. These differently expressed genes mainly involved in 148 functional categories and 61 pathways,including apoptosis,tumorogenesis,metabolism,signal transduction,cell cycle and material transportation. [Conclusion] Regulation of expressions of GADD45α,CDKN1A,CDKN2B,JUN,EGFR might be associated with the antitumor effect of wogonin.
