      摘 要:[目的] 通过观察Rac1shRNA对卵巢癌细胞Skov3侵袭能力的影响。[方法] Rac1shRNA载体 pGPU6/GFP/Rac1-524以脂质体LipofectamineTM 2000介导转染卵巢癌细胞株Skov3,Transwell侵袭实验检验转染细胞的迁移侵袭能力;扫描电镜观察转染Skov3细胞的超微结构变化。[结果] pGPU6/GFP/Rac1-524转染组细胞迁移距离最短,板状伪足消失,侵袭能力下降,与pGPU6/GFP/NC转染组和未转染组比,具有显著性差异(P<0.05)。[结论] Rac1shRNA载体pGPU6/GFP/Rac1-524通过使细胞伪足消失,可有效抑制卵巢癌细胞 Skov3的迁移侵袭。

      Abstract: [Purpose] To observe the influence of Rac1shRNA on invasive ability of ovarian cancer cell Skov3. [Methods]Rac1shRNA vector pGPU6/GFP/Rac1-524 mediated by liposome Lipofectamine TM 2000 was transfected to human ovarian cancer cell lines Skov3. Migration and invasion ability of transfected cell were detected by Transwell invasion test,and ultrastructural changes of Skov3 cells were observed by scanning electron microscopy.[Results]In pGPU6/GFP/Rac1-524 transfection group,the shortest migration distance of transfected cell,lamellipodia disappeared,invasive ability of transfected cell declined were found,those were significanty different between pGPU6/GFP/NC transfection group and the untransfection group.[Conclusion]Rac1shRNA vector pGPU6/GFP/Rac1-524 might effectively inhibit the migration and invasion of ovarian cancer cell Skov3 by making the cell pseudopod disappeared.
