      FOXO3a是Forkhead box O(FOXO)家族的重要转录因子,作为磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B(AKT)信号通路下游关键的转录因子,通过调节细胞周期抑制因子、促凋亡基因及抗氧化应激相关基因的转录,在维持细胞稳态、抑制肿瘤细胞异常增殖和调控细胞炎症反应中发挥关键作用。近年来大量研究表明,FOXO3a在不同肺癌组织病理类型的发生与发展过程中具有重要调控意义,FOXO3a普遍表现为抑制肿瘤生长、侵袭及转移的抑癌因子,其表达下调或功能失活与肿瘤进展、侵袭转移及治疗耐药性相关。本文将系统综述 FOXO3a参与调控非小细胞肺癌及小细胞肺癌发生发展的分子机制,为肿瘤疾病的诊断、预后评估及免疫靶向治疗提供新的理论依据和潜在分子靶点。
英文摘要:
      FOXO3a is an important transcription factor of the Forkhead box O (FOXO) family. As a key transcription factor downstream of the phosphatidylinositol 3-kinase (PI3K)/protein kinase B (AKT) signaling pathway, it regulates the transcription of cell cycle inhibitors, pro-apoptotic genes, and antioxidant stress-related genes, playing a crucial role in maintaining cellular homeostasis, inhibiting abnormal proliferation of tumor cells, and regulating cellular inflammatory responses. Recent studies have shown that FOXO3a has significant regulatory significance in the occurrence and development of different pathological types of lung cancer. FOXO3a generally acts as a tumor suppressor factor that inhibits tumor growth, invasion, and metastasis. Its expression downregulation or functional inactivation is associated with tumor progression, invasion, metastasis, and treatment resistance. This article will systematically review the molecular mechanisms by which FOXO3a regulates the occurrence and development of non-small cell lung cancer and small cell lung cancer, providing new theoretical basis and potential molecular targets for the diagnosis, prognosis assessment, and immunotargeted therapy of tumor diseases.
