      肝癌是世界第六大恶性肿瘤,发病率和死亡率都很高。其临床特点是起病隐匿、发展迅速、易转移、预后差,严重威胁着患者的健康。肝细胞癌(HCC)是原发性肝癌中最常见的类型,占所有肝癌病例的90%以上。目前,治疗HCC的主要方法是手术、放疗和化疗,但肿瘤的复发和转移仍然存在,而调控HCC发生和发展的机制研究尚不完善,给治疗方法和药物的创新带来了挑战。因此,需要探索更多调控HCC生长和转移的新机制。癌症干细胞(CSCs)是肿瘤中的一个小亚群,具有自我更新和无限增殖的特点。以往的研究表明,癌症干细胞具有高度的致瘤性、转移性和耐药性,并与肿瘤复发密切相关。肝癌干细胞(LCSCs)是导致 HCC的原因,也是其复发的根本原因。巨噬细胞是引起慢性炎症反应的最重要细胞。浸润肿瘤组织或肿瘤微环境(TME)的巨噬细胞称为肿瘤相关巨噬细胞(TAMs)。TAMs是肿瘤炎症微环境中最重要的组成部分,影响着HCC细胞的各种恶性行为和肿瘤内的免疫逃逸状态。TAMs是炎症微环境与癌细胞间的“桥梁”。以TAMs为靶点抑制LCSCs的形成可能为HCC的治疗提供一种可能性。本文综述了LCSCs的特点、在HCC中的主要作用、TAMs的亚型和功能、LCSCs与TAMs的相互关系,并简要总结了TAMs相关抗HCC治疗靶点的最新科研成果,为HCC的治疗提供新思路。

      Liver cancer is the sixth largest malignant tumor in the world, with high morbidity and mortality. Its clinical characteristics include latent onset, rapid development, easy metastasis, and poor prognosis, all of which seriously threaten patients’ health. Hepatocellular carcinoma (HCC) is the most common type of primary liver cancer, accounting for more than 90% of all liver cancer cases. At present, the main treatment methods for HCC are surgery, radiotherapy and chemotherapy, but tumor recurrence and metastasis still exist, and the research on the mechanism regulating the occurrence and development of HCC is not yet complete, bringing challenges to the innovation of treatment methods and drugs. Therefore, more new mechanisms that regulate the growth and metastasis of HCC need to be explored. Cancer stem cells (CSCs) are a small subset of tumors that have the characteristics of self-renewal and infinite proliferation. Previous studies have shown that CSCs have a high degree of tumorigenicity, metastasis, and drug resistance, and are closely related to tumor recurrence. Liver cancer stem cells (LCSCs) cause HCC and are the root cause of its recurrence. Macrophages are the most important cells causing chronic inflammatory reactions. Macrophages that infiltrating tumor tissues or tumor microenvironment (TME) are called tumor-associated macrophages (TAMs). TAMs are the most important component of the inflammatory microenvironment of tumors, affecting various malignant behaviors of HCC cells and the immune escape state within tumors. TAMs serve as a "bridge" between the inflammatory microenvironment and CSCs. Targeting TAMs to inhibit the formation of LCSCs may provide a possibility for HCC treatment. This article reviews the characteristics of LCSCs, their main role in HCC, the subtypes and functions of TAMs, and the interrelationship between LCSCs and TAMs, and briefly summarizes the latest scientific research results on TAMs-related anti-HCC therapeutic targets to provide new ideas for the treatment of HCC.
